肝细胞性肝癌中Caspase-3和PCNA的表达

目的探讨Caspase-3和PCNA在肝细胞肝癌的表达及其在肝细胞肝癌发生、发展中的作用。方法应用组织芯片技术和免疫组化方法（sP法）检测95例肝细胞肝癌组织、68例癌旁组织和19例正常肝组织中Caspase-3和PCNA的表达。结果Caspase-3在肝细胞肝癌中表达阳性率（34．7％）显著低于癌旁组织（79．4％，P〈0．01）和正常肝组织（84．2％，P〈0．01）；Caspase-3的表达与肝细胞肝癌的组织分级和HBsAg有关（P〈0．01，P〈0．05），与患者年龄、性别、肿瘤大小和转移无关（P〉0．05）。PCNA在肝细胞肝癌中的表达阳性率（68．4％）显著高于癌旁组织（26．5％，P〈0．01）和正常肝组织（15．8％，P〈0．01）；PCNA的表达与患者组织分级、转移有关（P〈0．01，P〈0．05），与年龄、性别、肿瘤大小、和HBsAg无关（P〉0．05）。在肝癌组织中Caspase-3与PCNA的表达呈显著负相关（r=-0．598，P〈0．001）。结论肝细胞肝癌中Caspase-3的低表达与PCNA的高表达在肿瘤的发生、发展过程中可能起重要作用。

Expression and clinical significance of caspase-3 and PCNA in hepatocellular carcinoma by the application of tissue microarray

Objective To investigate the clinical significance and effects of the expression of caspase - 3 and PCNA in the occurrence and development of hepatocellular carcinoma. Methods The expression of caspase - 3 and PCNA was detected by tissue microarray and immunohistochemical method in 95 hepatocellular carcinoma tissues ,68 parahepatic cancer tissues and 19 normal liver tissues. Results The expressions rate of caspase - 3 in hepatocellular carcinoma tissues （ 34. 7% ） was significantly lower than that of parahepatic cancer tissues （79.4%, P 〈 0.01 ） and normal liver tissues （84.2%, P 〈 0.01 ）. The expression of caspase -3 was correlated with Edmondson,s histological grade of carcinoma and HBsAg （P 〈 0. 01, P 〈 0.05 ）, but without correlation with patients, age, sex, diameter and metastasis （P 〉 0.05 ）. The expressions rate of PCNA in hepatocellular carcinoma tissues （68.4%） was significantly higher than that parahepatic cancer tissues （26.5%, P 〈 0.01, ） and normal liver tissues （ 15.8% , P 〈 0.01 ）, The expression of PCNA was correlated with Edmondson＇ s histological grade and metastasis of carcinoma （ P 〈 0, 01 ,P 〈 0.05 ）, but without correlation with age, sex, and HbsAg of the patients （P 〉 0.05） ; The expressions of Caspase-3 was negative correlated with PCNA（r = -0.598, P 〈 0.001）. Conclusion The low expression of caspase - 3 and a high expression of PCNA may play an important role in the curse of occurrence and progression of hepatocellular carcinoma.
