大孔吸附树脂分离纯化菊花中绿原酸工艺的研究

摘    要:目的研究从菊花中提取纯化绿原酸的最佳工艺。方法本文探索了不同型号的大孔吸附树脂静态吸附性能。以绿原酸的吸附率和解吸附率为指标,L9(34)正交设计法筛选绿原酸纯化条件。结果 AB-8大孔树脂对菊花中绿原酸分离纯化的综合性能较好。最佳纯化工艺为上样液绿原酸的质量浓度为1.8mg.mL-1,上样速度为3BV.h-1,柱径高比为1:8;最佳洗脱方式是:用2.5倍柱体积的水洗涤除杂,40%乙醇3BV洗脱。结论以上纯化绿原酸工艺稳定可行,可用于大规模制备。平均总绿原酸得率0.81%,含量52.43%。

Abstract:Objective To study the best method of purification of chlorogenic acid in Chrysanthemum morifolium Ramat.Methods Absorption and desorption prosperities of different macroporous resins were observed by static absorption method.With the reservation rate of chlorogenic acid as indexes,Orthogonal design with L9(34) was used to select condition of purification.Results Macroporous resin of AB-8 had the best separation and purification performance,and the optimum conditions were as follows: 40% alcohol and 2.5BV amount of solution,the flow rate was 3BV h-1.Conclusion The condition of purificationis was stable and practicable,it could be used for large scale preparation.The amount of chlorogenic acid extracted was 0.81% with a content of 52.43%.
