嗅鞘细胞移植治疗大鼠脑出血的实验研究

摘    要:目的:嗅鞘细胞移植入大鼠脑出血模型后,观察偏瘫大鼠功能恢复以及移植细胞后30 d内的形态学变化。方法:差速贴壁法培养出高纯度的嗅鞘细胞,制作38只大鼠脑出血模型,分三组:第1组10只为空白组,第2组10只为HBSS移植组,第3组18只为嗅鞘细胞移植组;三组大鼠分别在术后3 d、7 d、14 d和30 d用forelimb placing方法进行评分,判断嗅鞘细胞移植治疗大鼠脑出血模型的作用。结果:hoechst示踪及电镜观察结果,均证实嗅鞘细胞能在大鼠体内存活;对三组大鼠进行评分,嗅鞘细胞移植组的功能恢复明显优于其他两组。结论:从成年大鼠嗅黏膜用差速贴壁法能成功培养出嗅鞘细胞,并用于细胞移植,移植细胞能存活;嗅鞘细胞移植能改善大鼠脑出血后偏瘫肢体的功能。

Abstract:Objective: To transplant the successfully cultured olfactory ensheathing cells to rat cerebral hemorrhage mode and to observe the neurological repair function of OECs and the shape of cells in hematoma and brain tissue of 30 days. Methods: High purification of olfactory ensheathing cells using the method of the differing rates in attachment of the various cells type was cultured. Thirty-eight rats of cerebral hemorrhage mode were established and divided into 3 groups; the first group (10 rats) was the blank group ; the second group (10 rats) the HBSS transplanting group ; the third group (18 rats) the OECs transplanting group. The three groups were all scored in 3 days,7 days, 14 days and 30 days after operation by forelimb placing method. The contribution of OECs transplanton for curing rat cerebral hemorrhage with statistics method was eraluated, Results: OECs were comfirmed alive by immunofluorescence frace method and electron microscope; after scoring the three groups by forelimb placing method and calculating them, Olfactory ensheathing cells injected significantly reduced motor deficits compared with the control groups after cerebral hemorrhage (P〈0.05). Conclusion: OECs can be successfully cultrued using the method of the differing rates of attachment of the various cells type from adult mouse olfactory mucosa and can be used for cell transplanting; OECs transplanting method is a method of curing rat cerebral hemorrhage.
