摘 要: | 目的探讨丙戊酸(VPA)联合顺铂(DDP)对人肝癌细胞HepG2、BEC7404、SMMC7721的增殖抑制作用。方法三组浓度的VPA与DDP单药及联合作用于人肝癌细胞HepG2、BEC7404、SMMC7721,24、48、72 h后,观察细胞数量和形态的变化。用MTT法分析药物对细胞增殖的抑制率(IR),计算联合用药对细胞增殖抑制率q值,考察联合使用是否为协同作用。结果 72 h后,各用药组细胞数目减少十分明显,其中VPA+DDP组较单独用药组减少更为显著,其细胞形态发生较大改变。采用MTT比色法显示,对照组细胞生长率明显高于各实验组,联合用药组对细胞增殖IR更为显著,且随着药物浓度增加及作用时间延长,各组对细胞增殖IR明显增加。根据q值结果,在VPA较高浓度(≥100μg/mL)时,联合用药组具有明显的协同作用。结论 VPA能增强DDP对人肝癌细胞的增殖抑制作用,并且在VPA较高浓度时,两药具有协同优势。

Abstract: | Objective To investigate the proliferation inhibition effect of valproic acid (VPA)combined with cisplatin (DDP)on human hepatoma cell HepG2, BEC7404 and SMMC7721. Methods The single use and combination use of three sets of concentration of VPA and DDP were given to human hepatoma cell HepG2, BEC7404, SMMC7721. After 24,48 and 72 h, the changes in the number and morphology of cell was observed;and the cell prolif- eration inhibition rate(IR) of the drug was analyzed by MTF assay; the cell proliferation inhibition rate q value of combination therapy was calculated, and the synergistic effect whether used in combination was investigated. Results After 72 h, the number of cells in each treatment group decreased obviously, and the decrease of VPA + DDP group was more significant than that of single group, with great change in morphology. MTT assay showed that the cell growth rate of control group was significantiy higher than that of each experimental group; the VPA + DDP group had more significant effect on cell proliferation IR; with the increase in drug concentration and prolong duration of action, the proliferation IR of each group increased significantly. According to the q value in higher concentration of VPA(≥100 μg/mL) , VPA + DDP group had a significant synergistic effect. Conclusion VPA can enhance the proliferation of DDP on hu- man hepatoma cell, and they have a synergistic advantage when VPA in a higher concentration. |
