姜黄素对人鼻咽癌株CNE-2Z细胞凋亡的影响及机制研究

摘    要:
目的探讨姜黄素(CU)对人鼻咽癌株CNE-2Z细胞凋亡的影响及机制。方法体外培养CNE-2Z细胞在梯度浓度CU(0、20、40、80μmol/L)干预48 h后,采用MTT法检测其对CNE-2Z抑制增殖的作用,ELISA法检测8-羟基-2-脱氧鸟苷(8-OHdG)水平,Hoechst33342染色检测细胞形态学的变化,Western blotting法检测PI3K和Akt磷酸化水平。结果与空白对照组(0μmol/L)比较,CU干预后明显抑制CNE-2Z的细胞增殖(P<0.01),同时减少内源性的8-OHdG生成(P<0.01)。此外,CU干预可有效促进癌细胞凋亡,下调p-PI3K和p-Akt的表达。结论姜黄素具有良好的抗肿瘤作用,其机制与抑制癌细胞增殖、诱导细胞凋亡及抑制PI3K/Akt通路有关。

Abstract:
Objective To investigate the effect and mechanism of curcumin in the apoptosis of human naso- pharyngeal carcinoma cell line CNE-2Z. Methods After treatment of the stepwise concentrations of CU( 0,20,40,80 p.mol/L) in cultured CNE-2Z cells for 48 h,the anti-proliferation effect of the CU on CNE-2Z cells was assessed by using a MTT assay ; the level of 8-hydroxy-2-deoxyguanosine (8-OHdG)was detected by ELISA assay;the examination of morphological changes in CNE-2Z cells was observed by Hoechst33342 staining ; the phosphorylated level of PI3K and Akt was evaluated by Western blotting assay, respectively. Results Compared with the blank control group (0 txmol/L) ,CU treatment resulted in the marked anti-proliferative effect on CNE-2Z cells(P 〈0. 01 ) ,reduced endog- enous 8-OHdG generation ( P 〈 0. 01 ). In addition, CU treatment contributed to promoting apoptosis in cancer cells and decreasing the p-PI3K and p-Akt expressions( P 〈 0. 01 ). Conclusion Curcumin exerts the effective anti-tumor effect, and the mechanism is involved in inhibiting the cancer cell proliferation and inducing apoptosis, as well as the block of PI3 K/Akt pathway.
