Abstract:Objectives:To explore the protective effects and mechanism of Ophiopogon saponins D on hypoxia-reoxygenation injury of cardiomyocytes.Methods:Cardiomyocytes(H9c2)were cultured in vitro.Cells are divided into control(Con)group,hypoxiareoxygenation(H/R)group,hypoxia-reoxygenation+Ophiopogon saponin D(H/R+OpD)group,hypoxiareoxygenation+Ophiopogon saponin D+PI3K overexpression group(H/R+OpD+PI3K),hypoxia-reoxygenation+Ophiopogon saponin D+pcDNA empty vector(H/R+OpD+pcDNA)group.Thiazole blue(MTT)method was used to detect the survival rate of cardiomyocytes.Flow cytometry was used to detect cardiomyocyte apoptosis rate,malondialdehyde(MDA),lactate dehydrogenase(LDH)and superoxide dismutase(SOD)activity were detected by respective commercial detection kits.Enzyme-linked immunosorbent assay(ELISA)was used to determine the content of creatine kinase isoenzyme(CK-MB)and the content of inflammatory factors tumor necrosis factor-α(TNF-α)and interleukin 1β(IL-1β).Real-time fluorescent quantitative PCR(qPCR)and Western blotting were used to detect the mRNA and protein expressions of phosphatidylinositol kinase(PI3K)in cardiomyocytes of each group.Results:Compared with the Con group,the content of CK-MB,TNF-α,IL-1β,LDH and MDA,the apoptosis rate and the expression of PI3K in the cardiomyocytes of the H/R group were significantly increased,cell viability and SOD activity were significantly reduced(all P<0.05).Compared with the H/R group,these changes were significantly reversed in H/R+OpD group(all P<0.05).Compared with H/R+OpD group,the content of CK-MB,TNF-α,IL-1β,LDH and MDA,apoptosis rate were significantly increased,and the cell viability and SOD activity were significantly reduced in H/R+OpD+PI3K overexpression group(all P<0.05),while theses values were similar between H/R+OpD group and R+OpD+pcDNA group(all P>0.05).Conclusions:Ophiopogon saponin D possesses protective effects on myocardial cells undergoing hypoxia/reoxygenation injury via inhibiting PI3K gene expression.
