Abstract:
Objective To explore the effect of EZH2 on the migration and invasion of human cervical cancer cell lines and its mechanism.Methods CRISPR/Cas9 was used to knockout EZH2 expression in HeLa and SiHa cell lines.The migration was detected by scratch test,and the migration and invasion in vitro was detected by Transwell migration and invasion test.The STAT3 mRNA level was detected by real-time fluorescence quantitative PCR,and the expression of STAT3 and p-STAT3 proteins was detected by Western blot.ResultsCRISPR/Cas9 could knockout EZH2 expression.The wound healing rate of HeLa and SiHa cells in EZH2 knockout group at 24 h and 48 h,and the counting of transmembrane cell migration and invasion in Transwell chamber were lower than those in control group(P<0.05).Compared with the control group,the protein level of p-STAT3 in EZH2 knockout group was significantly decreased(P<0.05).Conclusions EZH2 plays an important role in promoting the migration and invasion of cervical cancer cells,which may be related to the targeted regulation of STAT3 pathway,and is a potential target in the treatment of cervical cancer.
