摘 要: | 目的通过E方法(效率法)相对定量动脉粥样硬化(AS)患者外周血白细胞基质金属蛋白酶9(MMP9)和基质金属蛋白酶组织抑制因子1(TIMP-1)基因表达及其临床意义。方法采用E法对动脉粥样硬化患者(男性61例,女性78例,平均年龄68岁)和健康体检者(男性19例,女性15例,平均年龄33岁)外周血白细胞MMP9和TIMP-1基因表达水平进行相对定量,数据通过SPSS 11.5统计软件进行分析,AS组与健康对照组间MMP9和TIMP-1基因表达水平差异显著性采用两样本t检验。结果 AS组TIMP-1表达水平则显著低于对照组,分别为0.78±0.06和1.14±0.26,差异具有统计学意义(P〈0.05)。而AS组MMP9表达水平为对照组的1.42倍,MMP9与TIMP-1的比值亦高于对照组,但差异均无统计学意义(P〉0.05)。结论与健康组比较,动脉粥样硬化患者外周血MMP9表达水平上调,而TIMP-1表达水平下调,动脉粥样硬化患者MMP9和TIMP-1之间存在调节失衡。

Abstract: | Objective to investigate the level of matrix metalloproteinase 9(MMP9) and tissue inhibitor of matrix metalloproteinase 1(TIMP-1) gene relative expression in peripheral blood leukocytes using E method(efficency method) and its clinical significance.Methods relatively to quantify expression of MMP9 and TIMP-1 in peripheral blood leukocytes in atherosclerotic(61 male and 78 female,average age of 68 years) and healthy volunteers(19 male and 15 female,average age of 33 years) using E-method.The data were analyzed for the significant difference of MMP9 and TIMP-1 gene expression level between AS group and healthy group by two-sample t test using SSPS 11.5 statistical software.Results The expression level of TIMP-1 in AS group was significantly lower than the control group(P0.05,respectively 0.78±0.06 and 1.14±0.26.The expression level of MMP9 in AS group was 1.42 times in the healthy group,the ratio of MMP9 and TIMP-1 also higher than the healthy group,but the differences ware not statistically significant(P0.05).Conclusion Expression of MMP9 in peripheral blood leukocytes of atherosclerosis patients was up-regulated,but expression of TIMP-1 was down-regulated compared with the healthy group,the imbalance between these enzymes takes an important role in the pathogenesis of atherosclerosis. |
