摘    要:目的观察三氧化二砷(AS_2O_3)对大肠癌细胞株SW620的生长抑制作用并初步探讨其机制.方法大肠癌细胞株SW620用不同浓度AS_2O_3处理.①MTT法观察不同浓度AS_2O_3作用不同时间后对SW620细胞生长的影响;②增殖细胞核抗原(PCNA)单抗LSAB免疫组化染色-流式细胞仪检测 PCNA阳性细胞百分比.结果①AS_2O_3明显抑制大肠癌细胞株SW620细胞增殖,其作用随AS_2O_3浓度增加和作用时间延长而增加;②随AS_2O_3浓度增加,PCNA阳性细胞百分比逐渐下降(p<0. 05)结论二氧化二砷可明显抑制大肠癌细胞株 SW620的生长,调控 DNA合成及细胞增殖的 PCNA蛋白合成降低可能是 AS_2O_3的作用机制之一.

Abstract:Objective To observe the effect of proliferation inhibition of Arsenic Trioxide (AS_2O_3) on colonic neoplasms cell line SW620 and its mechanism. Methods Colonic neoplasms cell line SW620 was treated with different concentration AS_2O_3. MTT assay was used to observe the proliferation status. Immunohis tochemistry staining method and flow cytometry were used to measure the percentage of proliferating cell nuclear antingen (PCNA) positive cell. Results AS_2O_3 could significantly inhibit the growth of SW620 cell, and this effect was dose and time dependent. The percentage of PCNA positive cell was decreased gradually when concentration of AS_2O_3 was increased. Conclusions Arsenic Trioxide could inhibit the proliferation activity of colonic neoplasms cell line SW620. Reduction of PCNA which was related to the regulation Of DNA synthesis and cell proliferation might be the mechanism of this proliferation inhibition.
