去甲肾上腺素和多巴胺对内毒素孵育兔肺动脉和体动脉环张力影响的比较

摘    要:目的探讨内毒素休克时不同浓度去甲肾上腺素(NE)和多巴胺(DOPA)对肺动脉、体动脉血管张力的影响。方法选用12只雄性健康大耳白兔。随机选取6只白兔制备离体肺动脉环和体动脉环,每只制备肺、体动脉环各6个,按肺、体动脉配对分为正常组和LPS组,予以NE干预,根据NE低、中、高剂量再分为三个亚组。其余6只白兔相同方法制备肺、体动脉血管环,亦配对分为正常组和LPS组,予以DOPA干预,根据DOPA低、中、高剂量再分为三个亚组。LPS组血管环用加入含4μg/mLLPS的RPMI营养液孵育建立内毒素休克模型,正常组血管环用RPMI营养液孵育。通过血管环张力测定观察血管环张力变化。结果①在正常组,低、中剂量NE和高剂量DOPA对体动脉的收缩作用均强于肺动脉(P均〈0.05),中剂量DOPA对体动脉收缩作用弱于肺动脉(P〈0.05),高剂量NE对体、肺动脉收缩作用无显著差异(P〉0.05),低剂量DOPA对体、肺动脉表现为舒张作用。②LPS孵育后,低剂量DOPA对体动脉的收缩作用弱于肺动脉(P〈0.05),低、中、高剂量NE和中、高剂量DOPA对体、肺动脉收缩作用均无显著差异(P均〉0.05)。③LPS孵育后,与正常对照组比较,NE对体、肺动脉收缩作用减弱或没有显著变化;中剂量DOPA对体动脉收缩作用增强(P〈0.05),对肺动脉收缩作用减弱(P〈0.05)。结论内毒组休克时,NE对体、肺动脉的收缩作用无显著差异,中剂量DOPA可使肺动脉收缩减弱而使体动脉收缩增强。
