APOA5-1131T/C基因多态性背景下糖尿病肾病同型半胱氨酸和脂蛋白a水平的相关性研究

摘    要:目的探讨载脂蛋白A5基因(APOA5)-1131T/C基因多态性与糖尿病肾病患者同型半胱氨酸和脂蛋白a水平的相关性。方法用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)技术检测40例健康人及45例糖尿病肾病患者APOA5-1131T/C基因型,并统计等位基因频率分布情况;用奥林帕斯2700型全自动生化分析仪检测同型半胱氨酸和脂蛋白a水平。结果糖尿病肾病组APOA5-1131T/C基因频率、等位基因频率与对照组差异无统计学意义(P〉0.05)。糖尿病肾病组C等位基因携带者同型半胱氨酸和脂蛋白a水平显著高于非C等位基因携带者(P〈0.05)。结论APOA5-1131T/C基因多态性影响糖尿病肾病患者同型半胱氨酸和脂蛋白a水平,C等位基因与糖尿病肾病同型半胱氨酸和脂蛋白a水平增高有密切关系。

Study on the Relationship of Apolipoprotein A5 Gene -1131T/C Polymorphism with lipid protein (a) and homocysteinemia levels in patients with diabetic nephropathy

Abstract:Objective To study the Relationship of Apolipoprotein A5 Gene -1131T/C Polymorphism with lipid protein (a) LP(a) ] and homocysteinemia(Hcy) levels in patients with diabetic nephropathy(DN). Methods The frequencies of genotypes and alleles of APOAS-1131T/C were determined in 45 DN patients by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and 40 healthy people were employed as controls. The levels of lipid protein (a) and homocysteinemia of all groups were also measured by Olympus AU 2700 analyzer. Results There was no significantly statistical difference of the frequencies of APOAS-1131T/C genotypes and alleles between DN patients and the control group(P 〉 0. 05 ). In DN group, the level of lipid protein (a) and homocysteinemia in C carriers (TC + CC) was significantly higher than that of non-C carriers (TT) (P 〈 0. 05 ). Conclusion APOAS-1131T/C gene polymorphism is associated with the level of lipid protein (a) and homocysteinemia in DN. APOAS-1131T/C alleles may lead to the increase of lipid protein (a) and homocysteinemia in DN.
