摘    要:目的建立不同剂型保健食品中维生素A、维生素A乙酸酯、维生素A棕榈酸酯、维生素E、维生素E乙酸酯和β-胡萝卜素的液相色谱检测方法。方法油状试样经乙酸乙酯提取,片剂和颗粒状样品试样经酶解后石油醚I提取,经高效液相色谱C18柱分离,甲醇-乙腈-水作为流动相,梯度洗脱,柱温箱为35℃,二极管阵列检测器检测,外标法定量。结果方法的线性关系良好,相关系数(r)均>0.999;检出限为0.05 mg/kg^2.00 mg/kg,定量限为2.5 mg/kg^100 mg/kg。相对标准偏差在1.2%~8.3%,回收率在83.7%~103%,稳定性满足实验要求。结论本方法通过改变样品的前处理方法,实现不同保健食品中6种脂溶性维生素的提取和净化,只测定维生素原形,无需皂化。该法适用于保健食品中脂溶性维生素的快速检测,方法简便、准确、重现性好。

Abstract:Objective The high performance liquid chromatography(HPLC)method was developed for the determination of fat-soluble vitamins in health food including all-trans-retinol,retinol acetate,ɑ-tocopherol,DL-alpha tocopherol andβ-carotene.Methods The vitamins in oil softgel samples were extracted with ethyl acetate,and those in tablets or granular samples were extracted with petroleum ether I after enzymolysis.The chromatographic separation was performed on a C18 column and detected with a PDA detector.The mobile phase consisted of methanol-acetonitrile-water and eluted in gradient elution mode,the column temperature was 35℃.The quantification was based on external standard method.Results Good linearity was observed with correlation coefficients above 0.999,the limits of detection were within 0.05 mg/kg-2.00 mg/kg,and limits of quantitation were within 2.5 mg/kg-100 mg/kg.The rates of recovery were within 83.7%-103%,and the RSDs were within 1.2%-8.3%.The method can be used for the qualitative and quantitative analysis of fat-soluble vitamins.Conclusion By changing the pretreatment method,the extraction and purification of six fat-soluble vitamins in different kinds of health food were realized.The original vitamins were determined without saponification by the method.The method is simple,accurate and good reproducibility for the determination of fat-soluble vitamins in health foods.
