药厂工作环境中细菌分布与16SrRNA测序鉴定

目的为了探究药厂工作环境中是否存在病原微生物污染。方法采集药厂工作环境可能存在的细菌样本,进行增菌培养后观察细菌菌落特征和显微镜下革兰染色特征。用水煮法提取分离出细菌的DNA,采用16SrRNA通用引物扩增出DNA片段,纯化PCR产物,统一送去华大基因公司测序,并进行拼接和校对DNA序列。在NCBI官方网站进行基因序列BLAST,鉴定细菌类别。结果 6个样品经分离培养后被鉴定为弯曲芽胞杆菌、巨大芽胞杆菌、短小芽胞杆菌、藤黄微球菌、科氏葡萄球菌、尿芽胞八叠球菌。结论该药厂工作环境中存在一定的病原微生物污染,应注意严格执行相应管理制度和卫生标准,保持工作环境符合无菌标准。

Bacterial distribution and the sequencing and identification of 16SrRNA in pharmaceutical environment

Objective In order to explore whether there is pathogenic microbial contamination in the pharmaceutical environment. Methods Colony characteristics and microscopic gram staining characteristics of bacteria were observed after collecting samples of bacteria that may exist in the working environment of pharmaceutical factory for enrichment culture. The DNA of the bacteria was extracted by boiling,and the DNA fragment was amplified by 16 SrRNA universal primers. The PCR product was purified and sent to Huada Gene Company for sequencing,and the DNA sequence was spliced and proofed. Gene sequence BLAST was conducted on the NCBI official website to identify bacterial species Results After isolation and culture,the samples were identified as Bacillus lentus,Bacillus megaterium,Bacillus pumilus,Micrococcus luteus,Staphylococcus aureus,and Staphylococcus urinaceus. Conclusion There is a certain pathogenic microbial contamination in the working environment of the pharmaceutical factory. It is necessary to strictly implement the corresponding management system and sanitary standards,and keep the working environment in compliance with the sterility standards.
