Bond Elut Mycotoxin固相萃取柱与免疫亲合柱在玉米油中玉米赤酶烯酮残留量液相色谱测定样品前处理中的性能比较

摘    要:目的比较Bond Elut Mycotoxin固相萃取与免疫亲合净化方法在高效液相色谱法测定玉米油中玉米赤酶烯酮残留量的优点与缺点。方法分别以Bond Elut Mycotoxin固相萃取与免疫亲合净化进行玉米油样品的处理,用高效液相色谱法(HPLC)测定玉米赤酶烯酮残留量,考察2种方法测定结果的相关性,比较2种方法的性能指标。结果 2种方法的定量结果差异无统计学意义(P>0.05);采用固相萃取净化法时,对于检出限≥8.0μg/kg的样品,2种方法的检出率基本相符;对于检出限<8.0μg/kg的样品,由于免疫亲合净化法进行了浓缩,其检出率高于固相萃取净化法。结论 2种方法均能应用于玉米油中玉米赤酶烯酮的测定。免疫亲合柱样前处理方法其处理过程繁杂,耗时,容易引入污染。而Bond Elut Mycotoxin固相萃取净化柱样品前处理方法简便,节约时间,提高通量,保质期长,更适用于大量样品的测定。

Abstract:Objective To compare the advantages and the disadvantages of Bond Elut Mycotoxin solid-phase extraction(SPE) and immunity affinity column(IAC) clean-up method for the determination of zearalenone residues in maize oil by High performance liquid chromatography(HPLC).Methods Maize oil samples were processed by Bond Elut Mycotoxin solid phase extraction and immunoaffinity purification.The zearalenone residues were determined by HPLC.The correlation between the results of the two methods was examined and the performance index of 2 methods was compared.Results The differences in the quantitative results of the two methods were not statistically significant(P>0.05);when the solid phase extraction purification method was used,the detection rates of the two methods for samples with a detection limit ≥8.0 μg/kg were basically consistent;for samples with a detection limit of<8.0 μg/kg,the detection rate was higher than that of solid-phase extraction purification due to the concentration by immunoaffinity purification.Conclusion Both the methods can be used for the determination of zearalenone in maize oil.IAC clean-up treatment process is complicated,time consuming and easy to introduce pollution.The Bond Elut Mycotoxin SPE clean-up treatment process is simple,rapid,and convenient,and can provide higher flux and longer shelf life.It is more suitable for the determination of a large number of samples.
