PCNA和COX-2在喉鳞状细胞癌中的表达及意义

摘    要:
目的探讨增殖细胞核抗原(PCNA)和环氧化酶-2(COX-2)在喉鳞状细胞癌组织中的表达及其与肿瘤发生和增殖的关系。方法用免疫组化SP法检测65例喉鳞癌组织及34例癌旁非肿瘤组织中PCNA和COX-2的表达,并分析两者的表达水平与各临床病理参数(年龄、吸烟、病程、饮酒、TN分期、病理分级)的关系。结果PCNA和COX-2在肿瘤组织中的阳性表达率分别为66.2%和72.3%,与对照癌旁非肿瘤组织比较(29.4%,20.6%)其表达显著增高,差别均有统计学意义。且PCNA和COX-2的表达相关。Logistic回归分析结果,t分期愈高、病程愈长易出现COX-2表达,淋巴结转移易出现PCNA表达。结论喉鳞癌组织中PCNA和COX-2的过度表达可能促进了喉鳞癌的发生发展,且两者在肿瘤的发生和发展中可能存在协同作用,PCNA和COX-2是喉鳞癌可能的治疗靶点。

The expression and significance of PCNA and COX-2 in laryngeal squamous cell carcinoma

Abstract:
Objective To investigate the expression of PCNA and COX-2 in laryngeal squamous cell carcinoma (LSCC) and the role in the tumorigenesis and proliferation of LSCC. Methods The expression of PCNA and COX-2 in 65 primary LSCC tissues and 34 adjacent non-neoplastic tissues were detected with immunohistochemical SP method. The relationship of PCNA and COX-2 and age, smoking, duration, alcohol consuming, TN staging, differentiation and clinical typing were studied. Results The expression of PCNA and COX-2 was observed in 66.2% and 72.3% of the case at the primary site and in 29.40% and 20.6 % of the adjacent non-plastic tissues, which demonstrated a significant difference from each other (P〈0.01). In addition,there was a significant correlation between the expression of PCNA and COX-2 (P〈0.01). The expression of PCNA and COX-2 in T3-4 stages LSCC was obviously higher than those in T1-2 stages(p 〈0. 05). The expression of PCNA in the Lymphaden transfered group was higher than that of the untransfered group(P 〈0.05), but the expression of COX-2 was not. There were no significant correlation between the expression of PCNA and cox-2 with other variables, including age, duration, smoking, alcohol consuming, differentiation and clinical typing. Conclusion The overexpression of COX-2 and PCNA was observed in LSCC tissues, which might play a crucial role in oneogenesis and proliferation of LSCC.
