摘    要:目的建立高效、灵敏的百草枯血浆浓度高效液相色谱检测法。方法流动相为乙腈-水(8:92),水相中含3.0 mmol.L-1辛烷磺酸钠、0.1 mol.L-1磷酸,二乙胺调pH值为3.0,流速为1.0 mL.min-1;检测波长258 nm;柱温28℃;内标物为Ethyl Viologen Dibromide。血浆沉淀蛋白处理后,进行高效液相色谱分析。结果色谱图基质干扰少,峰形好,线性范围0.1~10μg.mL-1,最低检测线0.01μg.mL-1,方法回收率96.65%~99.42%。结论高效液相色谱内标检测法适用于百草枯中毒患者的血药浓度检测。

Abstract:Objective To establish a high performance liquid chromatography (HPLC) method for the determination of paraquat in human plasma. Methods The mobile phase was acetonitrile - water ( 8 : 92), the water phase contained 3.0 mmol· L^-1 octane sulfonic acid sodium salt and 0.1 mmol· L^-1 phosphoric acid, adjusted pH to 3.0 with diethylamine, at a flow rate of 1.0 mmol· L^-1. The detector wavelength was 258nm,column temperature at 28 ℃. Using Ethyl Viologen Dibromide as internal standard. High performance liquid chromatographic analysis was carried out after plasma was precipitated. Results The chromatogram has few groundmass interference and good peak form. The detection limit of the method was 0.01 μg · mL^-1, and the average recoverie was 96.65% - 99.42%. Conclusion HPLC internal standard method can meet the requirement for analysis of paraquat in human plasma.
