肝细胞生长因子在肾脏缺血-再灌注损伤中的表达改变及意义

摘    要:目的通过对肾脏缺血再灌注时肾组织内肝细胞生长因子(HGF)mRNA及肾组织内丙二醛(MDA)和超氧化物歧化酶(SOD)活性的检测,揭示肾脏缺血再灌注过程中HGFmRNA表达变化及与SOD、MDA变化的相关性。方法雄性SD大鼠40只,切除右肾,1周后随机分为假手术组及实验组,每组20只,建立缺血再灌注损伤的模型。测定各组大鼠肾功能,对比2组间肾脏HGFmRNA、MDA及SOD的含量。结果实验组大鼠肾功能明显下降,表明肾脏缺血再灌注模型制作成功。与假手术组相比,缺血再灌注组肾组织SOD活性明显降低;MDA含量增高,二者相比差异具有统计学意义(P<0.05);肾脏HGFmRNA表达,再灌注组较假手术组增高,差异有统计学意义(P<0.01)。缺血再灌注过程中,肾脏HGFmRNA变化与SOD变化呈负相关,与MDA呈正相关。结论自由基的损伤性刺激可能是再灌注损伤过程中引起肾脏HGFmRNA表达上调的原因之一。

Abstract:Objective To investigate the mRNA expression of hepatitis growth factor (HGF) and the contents of SOD and MDA in kidney after renal ischemia-reperfusion injury (IRI) for a probable correlation between HGF mRNA expression and SOD, MDA contents during IRI process. Methods A total of 40 male SD rats received right nephrectomy and were randomly divided into the sham group and the experimental group at one week later. Renal IRI model was established by clasping left renal artery. Renal function after IRI was investigated by measuring serum creatinine (SCr) and blood urea nitrogen (BUN). Renal mRNA expression of HGF was detected by RT-PCR. Renal contents of SOD and MDA were analyzed by chromatometry. Results Rats in IRI group demonstrated deteriorated renal function compared with those in the sham group. Renal IRI induced a significant up-regulation of HGF expression (P〈0.01), increase in MDA (P〈0.05) and reduction in SOD (P〈0.05). The expression of HGF was positively correlated with SOD and negatively correlated with MDA. Conclusion Up-regulated expression of HGF mRNA during IRI may result from traumatic stimulation induced by free radicals.
