霉酚酸酯对糖尿病大鼠肾组织早期炎症反应的影响

摘    要:目的:探讨霉酚酸酯(MMF)对链脲佐菌素(STZ)诱导的糖尿病大鼠肾组织早期炎症反应的影响.方法:36只雄性SD大鼠随机分为正常对照组、糖尿病模型组和MMF治疗组,于4周、14周末每组分别处死6只大鼠,处死前进行24 h尿蛋白定量检测,处死后免疫组织化学染色观察调节激活正常T细胞表达和分泌的细胞分子(RANTES)、单核/巨噬细胞表面特异性标志抗原(ED-1)、Ⅳ型胶原蛋白(Col-Ⅳ)在肾组织中的表达,RT-PCR方法观察RANTES mRNA在肾组织中的表达.结果:MMF可显著抑制大鼠24 h尿总蛋白的排泄,下调肾组织RANTES,ED-1,Col-Ⅳ蛋白及RANTES mRNA的表达.结论:早期应用MMF对糖尿病大鼠的肾保护作用可能与抑制肾脏早期炎症反应有关.

Abstract:Objective To investigate the effect of mycophenolate mofetil(MMF) on early inflammatory reaction of renal lesion in streptozotocin(STZ)-induced diabetic rats.Methods Thirty-six male Sprague-Dawley rats were randomly divided into 3 groups after uninephrectomy: normal control group,diabetic model group,and MMF-treated group.Six rats in each group were sacrificed at the 4th week and 14th week after STZ injection.Twenty-four hour urinary protein(24 h Upro) count was measured before death.The expressions of regu...
