左旋四氢帕马汀对异丙肾上腺素致大鼠急性心肌缺血损伤的保护作用

摘    要: 目的研究左旋四氢帕马汀对异丙肾上腺素(ISO)致大鼠急性心肌缺血的保护作用及机制。方法采用大剂量ISO sc 诱导产生大鼠急性心肌缺血模型(ISO 30 mg·kg-1,每日1次,连续2d),实验大鼠随机分为正常组、ISO模型组、阳性时照组、左旋四氢帕马汀(60,120mg·kg-1)给药组,测定心脏系数和血清中乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)和丙二醛(MDA)含量。观察心肌组织谷胱甘肽过氧化物酶(CSH-Px);Na+-K+-ATPase,ca2+-Mg2+-ATPase活性。观察左旋四氢帕马汀的保护作用。结果左旋四氢帕马汀能显著降低血清中CK和LDH的释放;也能显著降低血清和心脏匀浆中MDA水平,心脏重量参数显示对心肌水肿有明显的抑制作用;保护心肌GSH-Px,Na+-K+-ATPase和Ca2+-Mg2+-ATPase活性。结论左旋四氢帕马汀对ISO所致大鼠急性心肌缺血有显著的保护作用。

Abstract:OBJECTIVE To study the protective effect of L-tetrahydropalmatine(THP)on myocardial ischemia induced by isoproterenol (ISO) .METHODS Myocardial ischemia rats were induced by subcutaneous injection of ISO (30 mg ·kg-1 for two days,once a day) . Rats were randomly divided into the blank control group, ISO group, positive control group and two THP groups (60,120 mg·kg-1). Cardiac indexes were examined. The levels of LDH,CK,MDA in serum were measured. The contents of MDA,GSH-Px and ATPase activity in heart were assayed by colorimetric analysis. RESULTS Compared with the blank control group, THP significantly reduced the cardiac indexes, obviously decreased the level of MDA, LDH and CK in the serum. THP significantly increased the activities of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and GSH-Px. CONCLUSION THP alleviated the acute myocardial ischemia injury induced by ISO in rats.
