外周血内皮祖细胞数量及eNOS基因表达在动脉粥样硬化大鼠中的改变

摘    要:
目的:探讨外周血内皮祖细胞(EPCs)数量及动脉内皮细胞型一氧化氮合酶(eNOS)基因表达在动脉粥样硬化(AS)大鼠中的改变。方法:将70只大鼠分为3组:正常对照组20只,采用假手术+普通饲料喂养;实验对照组20只,采用假手术+高脂饲料喂养;AS组30只,采用球囊损伤主动脉内皮、维生素D33×105 U·kg-1·d-1分6次肌内注射和饲以高脂饲料喂养。90d后分别采外周血进行EPCs的分离培养,于倒置相差显微镜下计数内皮祖细胞克隆形成单位(EPC-CFU),并采用Real-time PCR法检测腹主动脉中eNOS基因的表达。结果:与正常对照组和实验对照组相比,AS组外周血EPCs数量明显减少(P<0.01),eNOS基因表达明显下调(P<0.01);EPCs数量及eNOS表达在正常对照组和实验对照组中差异无统计学意义。结论:AS时EPCs数量减少,eNOS基因表达明显下调。

Abstract:
Objective:To analyse the change of the quantity of the endothelial progenitor cells(EPCs) from peripheral blood and the expression of endothelial nitric oxide synthase(eNOS) in atherosclerotic rats. Method:Seventy rats were divided into 3 groups: group 1(n=20) were fed with normal diet,group 2(n=20) were sham-operated and fed with high-fat diet,group 3(n=30) were injured by balloon in the endothelium of the arteries,and intramuscular injected with vitamin D3(3×105 U·kg-1·d-1) followed by high-fat diet.Peripheral blood samples were drawn to isolate and culture EPCs respectively,EPCs populations were assessed by the colony forming unit assay(EPC-CFU) through an inverted phase contrast microscope after 14 days culture,and genetic content of eNOS was detected by Real-time PCR 90 days later. Result:Compared with group 1 and group 2,the number of EPCs in group 3 group was lower(P<0.01),and group 3 had less expression of eNOS(P<0.01);but these values did not have significant difference between group 1 and group 2. Conclusion:Circulating EPCs populations are decreased,and the expression of eNOS gene is down-regulation in atherosclerotic rats.
