人参皂甙Rg3对肝癌细胞移植瘤模型的作用研究

摘    要:目的观察人参皂甙Rg3(简称Rg3)对裸鼠肝癌移植瘤模型的作用。方法采用人肝癌Bel-7402细胞株种植于12只雄性裸鼠左前上肢腋窝皮下,随机分成两组(n=6),Rg3组及对照组。实验自肿瘤接种第3天开始,人参皂甙Rg3(10mg/kg)隔日灌胃,共20次。停药1周后,脱颈处死,分别观察裸鼠的生存质量(包括精神状态、活动状况、对刺激的反应、体质量下降幅度、食欲和尿、便6项指标),观察检测肿瘤的重量,并计数肿瘤内微血管密度(MVD),观察瘤组织的Bax、Bcl-2的表达。结果Rg3组对肝癌裸鼠种植肿瘤的生长有明显的抑制作用,Rg3组MVD明显低于对照组,Rg3组Bax明显高于对照组、Bcl-2明显低于对照组。结论Rg3明显抑制肿瘤新生血管形成,明显降低肿瘤内的MVD,提高Bax、降低Bcl-2表达,抑制裸鼠肝癌移植瘤的生长,改善荷瘤生存质量。

Abstract:Objective To study the effect of ginsenoside Rg3(Rg3) on tra nsplanted human hepatic carcinoma in nude mice. Methods 12 male nude mice were randomly divided into 2 groups (n=6) to Rg3 (10 mg/kg) group and control group after being subcutaneously transplanted with vitro cultured Bel-7402 cell strain at the of armpit in left anterior-superior limbs. 3 days after being inoculated in nude mice, Rg3 was given by gastrogavage once every 2 days, for total 20 times. A week after treatment being stopped, mice were killed, the quality of life (including mental state, activity, response to stimulus, decrease of body mass, appetite, urine and stool)were observed in nude rats, and the sizes of solid tumors were measured, the intratumor MVD was examined by immunohistochemical staining. The expression of Bax, Bcl-2 protein of transplanted human hepatic carcinoma in nude mice was observed. Results The tumor weight of Rg3 group was significantly lower than that of control group. The MVD value of Rg3 group was significantly lower than that of control group. The expression of Bax of Rg3 group was lower than control group, while Bcl-2 of Rg3 group higher than control group. Conclusion Rg3 can obviously reduce the intratumoral MVD through inhibiting aniogensis of malignant tumor. Rg3 can lay down the expression of Bcl-2 while amplify the expression of Bax. The Rg3 can significantly inhibit the growth of transplanted human hepatic carcinoma as well as ameliorate the life quality in nude mice.
