氟中毒大鼠睾丸细胞DNA和RNA变化的荧光组织化学研究

摘    要:目的:探讨氟中毒对睾丸结构和功能损害的分子机制,研究其睾丸DNA和RNA的影响。方法:选用2月龄Wistar雄性大鼠16只,均分为实验组和对照组,实验组饮用含200mg/L氟化钠的去离子水,对照组单纯饮用去离子水。12周后,采用吖啶橙荧光染色法显示睾丸细胞DNA和RNA。结果:与对照组相比,实验组大鼠睾丸细胞DNA和RNA荧光显色反应强度明显减弱。结论:氟中毒大鼠睾丸细胞DNA和RNA活性明显减少,这些变化可能是氟损害睾丸结构和功能的重要原因之一。

Abstract:Objective:To study the changes of DNA and RNA content in testivular cells of fluorosis rat.Methods:Fluorosis rat was prepared by using NaF solution (200mg/L)as drinking water ad lib to Hale Wistar rat for 12 weeks.DNA and RNA content in testicular cells were defected for NaF fed and control rats by acridine orange(AO)staining and examined with fluorescent microscopy. Results:The DNA and RNA AO fluorescent staining density in the NaF treated group as markedly decreased as compared with the control group.Conclusion:Fluorosis may decrease the DNA and RNA content,which may be one of the molecular mechanism of the injurious action of fluorosis on the development and function of testis.
