消瘀散巴布剂和湿敷贴剂外用治疗急性软组织损伤的实验研究

摘    要:
目的:观察和比较消瘀散新旧剂型局部外用对软组织损伤(急性血瘀症)模型新西兰大白兔的治疗效果和透皮性能,以选择优势剂型。方法:将60只新西兰大白兔打击造成血瘀证模型后,随机分为模型组(A)、软膏剂组(B)、巴布剂组(C)、湿敷贴剂组(D);A组予空白贴剂外敷,C、D组及B组分别用消瘀散新旧剂型治疗。用药后第2、4、6天作大体观察评分、苏木精-伊红染色光镜下组织学观察及高效液相色谱仪测定大黄素血药浓度。结果:大体观察评分,D组与B组第4、6天比较,C组与B组第6天比较,症状有明显改善(P〈0.05,P〈0.01);组织病理革兰氏染色可见,第4天组织学观察评分D组指标明显优于B组(P〈0.01);血药浓度测定显示,第2、4、6天新剂型组(C组与D组)与旧剂型组(B组)比较差异有统计学意义(P〈0.01)。结论:消瘀散外用治疗急性血瘀证模型新西兰大白兔,新剂型的吸收和疗效优于旧剂型,新剂型中的湿敷贴剂疗效略优于巴布剂。

Abstract:
Objective: To observe and compare the therapeutic efficacy and the transdermal performance of different formulations of Xiaoyusan for external use in treating New Zealand White rabbit models with acute injury of soft tissue,and to select the better formulation.Methods:Models of blood stasis were established in all of 60 New Zealand White rabbits,and then they were randomly divided into 4 groups: the model group(A),treated with empty one;ointment group(B),treated with the old formulation;cataplasm group(C),treated with the new formulation;wet compress patch group(D),treated with the new formulation.Each rabbit was treated respectively with different formulations of Xiaoyusan for 2,4 and 6 days,general observation were taken and scored;HE staining were observed under light microscope;the plasma concentration of emodin were determined by the high performance liquid chromatographic(HPLC).Results:There was difference in general observation between group D and group B in the 4th 、6th day;there was difference in general observation between group C and group B in the 6th day(P<0.05,P<0.01);the score of pathological tissue section in group D was higher than that in group B in the 4th day(P<0.01);there was significant differences in blood concentration between the new formulation groups(C group and D group) and the old formulation group(B group) in the 2nd,4th,6th days(P<0.01).Conclusion:The absorbability and curative effect of Xiaoyusan new formulation for external use in treating the New Zealand White rabbit models with acute blood stasis was better than that for external use with old formulation;the curative effect of wet compress patch was a few better than cataplasm in new formulation.
