超短波对大鼠视神经挫伤后视网膜神经节细胞和BCL-2/Bax表达的影响

摘    要:
目的 研究大鼠视神经夹伤后,经定期超短波治疗视网膜神经节细胞(RGCs)数量的改变和Bcl-2/Bax蛋白表达的变化.方法 建立大鼠神经夹挫伤动物模型,随机选取5只大鼠作为正常组;选取30只作为实验组,伤后即开始超短波治疗;另外选取30只作为对照组,不接受超短波治疗,二者分别于伤后3d、5d、7d、10d、14d、28d处死.HE染色观察RGCs的动态变化,免疫组化方法检测RGCs表达Bcl-2及Bax的水平.结果 对照组视神经损伤后3d RGCs数目严重下降,2周内RGCs快速减少,2周以后继续减少;实验组神经损伤后3d RGC数目急剧下降,但之后各组RGCs数量没有明显降低;伤后在实验组和对照组中Bcl-2和Bax表达随访后时间延长都有不同程度的减少,Bcl-2/Bax的比值在实验组逐渐升高,对照组无明显升高.结论 神经损伤后RGCs数目减少是其视功能下降的病理基础之一,超短波治疗可提高RGCs的存活率,Bcl-2/Bax在RGCs死亡机制中起重要作用,Bcl-2/Bax比率与RGCs的数目变化有一定程度的相关性.

Abstract:
Objective To investigate the effect of ultrashort wave therapy after optic nerve crush and the change of the number of RGC and the express of Bcl-2/Bax in rat.Methods Produce optic nerve crush model in rats, half of them as experimental group, and start to give ultrashort wave therapy immediately.The others as control group, and they do not give ultrashort wave therapy.All of the rats are executed at 3d, 5d, 7d, 10d, 14d and 28d after the crush.And select 5 rats as normal group.Changes of retinal ganglion cells (RGCs) were observed by microscopic examination of the hemtoxylin and eosin (H&E) stained slides.Bcl-2 and Bax expressions in the retina were studied by immunohistocemistry and imaging analysis technique,Resuits Three days after the crush, the number of RGC in experimental group declined sharply.But the number of RGC in both groups did not reduce significantly.The expression of Bcl-2 and Bax had different degree of reduction after the injury in both experimental and control groups.The ratio of Bcl-2/Bax gradually in creasedin experimental group, but there is no significant increase in control group.Conclusions The decline of the number of RGC is one of the pathological bases in the decline of visual function.Ultrashort wave therapy can improve the survival of RGC, Bcl-2/Bax play an important role in the death mechanism of RGC; the ratio of Bcl-2/Bax has a certain degree correlation with the change of the RGC number.
