少阳生骨方对大鼠在体软骨损伤关节液IL-1β及软骨修复组织Ⅱ型胶原影响的实验研究

摘    要:目的应用少阳生骨方对大鼠在体软骨损伤进行干预,观察其对大鼠在体软骨损伤关节液中IL-1β水平及和修复组织中Ⅱ型胶原的影响。方法选用在相同条件下喂养的6~8周清洁级SD大鼠,制作成关节软骨损伤动物模型,按照组内均衡一致原则随机编为3组,即少阳生骨方组实验组(S组)、盐酸氨基葡萄糖胶囊组对照组(A组)和生理盐水空白对照组(K组),每组9只。于造模完成后的第二天,开始灌胃药物,S组少阳生骨方灌胃,A组盐酸氨基葡萄糖胶囊灌胃,K组生理盐水灌胃。对比观察少阳生骨方及盐酸氨基葡萄糖胶囊对大鼠关节软骨损伤后不同时期(4周、8周、12周)关节液中IL-1β水平和修复组织中Ⅱ型胶原的影响。结果三组关节液中IL-1β水平检测,S组和A比较:第4、8、12周A组高于S组(P〈0.05);第4、8、12周K组高于S组(P〈0.05);第4、8、12周K组高于A组(P〈0.05)。治疗前后自身比较:S组第8周、第12周均低于第4周(P〈0.05);A组第8周、第12周均低于第4周(P〈O.05);K组第4周〉第8周〉第12周(P〈0.05)。组织学观察显示S、A组软骨缺损修复组织的数量及质量均优于K组,S组的修复效果强于A组。结论通过和解少阳的方法可显著降低关节液中导致软骨细胞损伤的炎性介质IL-1β水平,提高软骨损伤后修复组织中Ⅱ型胶原的表达,从而有助于软骨损伤的修复。

Experimental study on the effect of Shao-yang formula in vivo cartilage damage in rats
Abstract:Objective To observe the repairment of articular cartilage injury and confirm its repairment of cartilage defects treated by Shao-yang formula. Methods 6 to 8 Weeks pure SD rats were randomly assigned into Shao-yang group(S group, n=9), controlling group (A group, n=9) and, blank group (K group, n=9). All three groups were fed in the same conditions (SD rats Provided by Luzhou Medical College Experimental Animal Center ). The day after the completion of the modeling, the S group were gavaged with Shao-yang formula, A group with Glucosamine Hydrochloride Capsules and K group with saline. The level of IL-1β in joint fluid in all three groups were assayed at the 4th weeks, 8th weeks and the 12th weeks. Results At the 4th, 8th and the 12th weeks, the level of IL-1β in group A was significantly higher than the that of S group (P〈O.05); the K group was higher than the that of S group (P〈O.05) and the K group was higher than the that of A group (P〈O.05). The level of 1L-1β at the 8th and 12th week were significantly less than that of the 4th week in S group and in A group respectively. In K group, the level of IL-1β at the 4th week was significantly higher than that of 8th and 12th week(P〈 0.05). Conclusions Shao-yang formula is helpful to the repairment of articular cartilage injury due to its reducement of IL-1β level in joint fluid.
