碱性成纤维细胞生长因子及其受体与肺癌新生血管(英文)

摘    要:目的探讨碱性成纤维细胞生长因子及其受体促进肺癌新生血管形成的机制。方法收集56份原发性支气管肺癌的标本,采用免疫组化 SABC 法检测微血管密度(MVD)、碱性成纤维细胞生长因子(bFGF)及其受体(FGFR-1)在肺癌组织中的表达状况.用 Kaplan-Meier 方法比较高、低血管密度组的生存时间。结果 (1)肿瘤组织 MVD 与患者的性别、年龄、病理类型、T 分期、M 分期无相关性,与 N 分期(P<0.01)和临床分期(P<0.05)关系密切。(2)高血管密度组的患者生存时间短,预后差(P<0.01)。(3)肺癌组织中的 bFGF 与 FGFR-1的表达也同 N 分期(P<0.05)和临床分期(P<0.05)相关。(4)肺癌组织中的 bFGF 表达在低血管密度组和高密度组的表达有显著性差异(P<0.01),FGFR-1在不同血管密度组的表达差异不明显。(5)肺癌组织中 bFGF 与 FGFR-1的表达具有一致性。结论(1)肺癌组织的微血管密度和 bFGF 的表达可以作为评估疗效、推测预后的指标。(2)bFGF 和 FGFR-1结合后,对肺癌新生血管的形成具有促进作用。

Abstract:Objective:To study the relationship between angiogenesis and the expression of bFGF and FGFR-1 in hmg cancer.Methods:The specimens of 56 patients with lung cancer treated with surgery were collected.Anti-Von Willebrand factor antibody was used to measure microvascular density (MVD) by means of SABC immunohistochemical technique,and antibody to basic fibroblast growth factor (bFGF) and its receptor (FGFR-1) to detect the expression of these three proteins in the tumor tissues.The survival time was compared between low MVD and high MVD groups by the Kaplan-Meier method.Results:(1) The expression of MVD showed no significant difference in some clinical characteristics,including sex, age,T stage,M stage and pathologic type,but significant difference in N stage (P<0.01) and clinical stage (P<0.05).(2) Survival analysis showed that high MVD group was associated with a risk of death (P<0.01).(3) The expression of bFGF and FGFR-1 were both related to lymphatic metastasis and clinical staging (P<0.05).(4) Significant difference was seen between low MVD and high MVD groups in the bFGF expression in lung cancer (P<0.01),whereas no correlation in FGFR-1.(5) High co-expression of bFGF and FGFR-1 was consistent in tumor cells.Conclusion:(1) MVD is a good prognostic factor for patients of lung cancer,and the same as bFGF.(2) The angiogenesis may be induced after bFGF binding to FGFR-1.
