脑神经细胞膜脂流动性的荧光偏振测定法

摘    要:以大鼠大脑神经细胞为实验材料,探索用荧光偏振法测定完整神经细胞的膜脂流动性、荧光探针DPH(1,6-Diphenyl-1,3,5-Hexatriene)标记的实验条件。适合条件为DPH浓度2×10-6mol/L,细胞浓度1×1010个/L,标记温度25℃,时间30min。本文还对无血清体外培养的人胚大脑神经细胞的膜脂流动性进行了初步动态观察,发现神经细胞在培养至3、6、9和12天时,膜脂流动性维持在相对稳定的水平。

Abstract:The membrane lipid fluidity of brain neural cells labeled with fluorescence probe 1,6-diphenyl-1,3,5-hexatriene (DPH) was determined by fluorescence polarization. The brain neural cells of newborn rat were used as experimental materials. The appropriate conditions for DPH labeling were: DPH concentration at 2 ×106 mol/L and cell concentration at 1010 cell/L kept in a water bath at 25℃ for 30 mimutes, A preliminary experiment was also carried out on the membrane lipid fluidity of neural cells from human fetal brains. The results indicated that the membrane lipid fluidity of cells after cultured for 3,6, 9 and 12 days tended to maintain on relative stable levels.
