不同月龄雌性大鼠骨质疏松模型研究

摘    要:
目的:观察不同月龄大鼠去卵巢后体重、骨矿含量(BMC)、骨密度(BMD)的变化,以确定骨质疏松模型形成的时间。方法:将Wistar雌性大鼠分成正常、2月龄、3月龄和8月龄4个组。正常组作为确定手术月龄的参考。对2、3、8月龄大鼠分别进行卵巢去除和伪手术。全部大鼠麻醉后在美国Lunar公司DPXL型骨密度仪上定期进行骨矿测量。测量前大鼠称重、测量后分析BMC、BMD随时间的变化规律以及卵巢去除与伪手术组之间的差异。结果:不同月龄施行卵巢去除BMC、BMD变化规律不同。2月龄大鼠去势后,4个月内2组的测量值没有统计学差异,均呈现增长趋势。3月龄大鼠去势2个月后BMC、BMD基本稳定,且测定值明显低于伪手术组。8月龄大鼠手术1周后,2组显示差异。结论:大鼠去除卵巢的月龄对骨质疏松造模有重要影响。本实验为去势大鼠造模提供可靠的依据。

Abstract:
Objective:To determine the time of formation of osteoporosis model on the basis of changes of body weight,BMC and BMD of rats of different month old.Methods:Wistar female rats were divided into normal and operation groups.The two,three and eight month old rats were separately operated by bilateral and sham ovariectomy.After general anesthesia,all the rats were periodically measured by Dual energy X ray Absorptiometry(DXA) of Lunar DPX L model and weighing.After measurement,the BMC and BMD at lumbar and femur were analyzed and compared between ovariectomy and control groups.Results:the weights,BMC and BMD of young rats quickly increased,while those of the rats over three month old increased slowly and those of eight month old rats were basically stable.Between ovariectomy and sham groups,BMC and BMD of two month old rats were of no significant difference.The BMC and BMD of ovariectomy group of three months old rats were significant lower than those of sham group.Conclusion:Age difference of ovariectomy ratshas important influence on osteoporosis model.
