肾综合征出血热病毒R22株感染人血管内皮细胞的核酸原位杂交研究

用地高辛配基（Ｄｉｇ－）标记我国肾综合征出血热（ＨＦＲＳ）病毒Ｒ２２株片段ｃＤＮＡＲ３克隆探针对ＨＦＲＳ病毒Ｒ２２株感染的人血管内皮细胞（ＨＥＣｓ）作核酸原位杂交，杂交结果显示在受染的血管内皮细胞胞浆内有明显的杂交信号，而正常细胞和ＨＦＲＳ病毒感染经ＲＮＡ酶处理组不出杂交信号，结果表明ＨＦＲＳ病毒ＲＮＡ主要定位于血管内皮细胞的细胞浆中，提示病毒ＲＮＡ在胞浆中复制。

In situ hybridization of nudeic acid in human vascular endothelial cells infected by hemmorrhagic fever with renal syndrome virus

RNA genome of R 22 strain of hemorrhagic fever with renal syndrome (HFRS) virus was detected in human vascular endothelial cells (HVECs) with in situ hybridization using Dig labelled R 3 cDNA probe. The positive hybridization signal was seen in the cytoplasma of the infected HVECs but not in the control cells including the normal HVECs and the infected HVECs pretreated with Rnase. This indicates that RNA of HFRS virus is mainly located and replicates in cytoplasma of HVECs.
