| 摘 要： | 通过ＤＮＡ重组技术和ＰＣＲ技术，将Ｆａｂ表达载体改建成了ＳｃＦｖ表达载体。该载体可以表达表达方式产生噬菌体抗体，用于抗体库技术，也可以分泌型表达产生有活性的ＳｃＦｖ。所表达的抗ＨＢｓ ＳｃＦｖ具有与其亲本Ｆａｂ相同的抗体活性。载体上具有聚组氨酸编码序列，使表达的ＳｃＦｖ可经金属螯合层析法进行简便有效的纯化。 |

| Abstract: | ScFv expression vectors were constructed from a Fab expressing vector by DNA recombination and PCR techniques.The vectors could express both phage surface displayed and soluble secreted ScFv,thus could be used in phage antibody library technology.The expressed ScFv showed same specificity against HBsAg as its parent Fab.A poly histidine attached to the secreted ScFv facilitated the purification of the ScFv by metal chelated chromatography. |
