摘    要:目的 研究ketanserin对慢性心肌梗死大鼠血流动力学的影响。方法 设假手术组、心肌梗死组和心肌梗死治疗组。治疗组在冠脉结扎后饮水中给予ketanserin 10mg·kg-1·d-1,持续 4~ 5wk。在清醒自由活动条件下连续监测血压 ,并采用改良Symth法测定动脉压力感受性反射敏感性(BRS)。结果 与假手术组比较 ,心肌梗死大鼠血压下降、心动间期波动性 (HPV )升高、BRS降低 ,而血压波动性(BPV)和心动间期无明显改变。ketanserin长期治疗可降低心肌梗死大鼠的血压、BPV和改善BRS。心肌梗死组室性心律失常发生率为 60 % (12 / 2 0 ) ,经ketanserin治疗后降至2 8 6% (6/ 2 1)。治疗或未治疗心肌梗死大鼠如果存在心律失常 ,则HPV升高 ,否则正常。结论 ketanserin可能作为改善心肌梗死预后的治疗药物。心肌梗死大鼠HPV增加与心律失常有关

Abstract:AIM To study the effects of ketanserin on haemodynamics in rats with chronic myocardial infarction (MI). METHODS Rats were divided into sham operated (Sham), MI and ketanserin treated MI groups. In treatment group, after MI with ligation of the left anterior descending artery, ketanerin was given through drinking water for 4~5 weeks. Blood pressure (BP) was continuously recorded in conscious unrestrained rats. Baroreflex sensitivity (BRS) was measured by modified Symths method. RESULTS In chronic MI rats, BP was decreased, heart period variability (HPV) increased, and BRS impaired. However, there were no significant changes in BP variability (BPV) and heart period. In ketanserin treated MI rats, both BP and BPV were reduced, and BRS was improved, when compared with untreated MI rats. The occurrence of ventricular arrhythmia was higher in untreated (12/20, 60%) than ketanserin treated (6/21, 28.6%) MI rats. Either in untreated MI rats or in ketanserin treated MI rats, if there was arrhythmia, HPV was larger. If not, HPV was normal, similar to the Sham levels. CONCLUSION Ketanserin may be as a new drug for improvement of MI prognosis. Large HPV in chronic MI rats is associated with arrhythmia.
