| 摘 要： | 目的建立能够准确反映注射用尖吻蝮蛇血凝酶蛋白质含量的测定方法。方法采用Lowry法、Brad-ford法、RP-HPLC法及AQC柱前衍生HPLC法四种方法测定注射用尖吻蝮蛇血凝酶蛋白质含量并进行比较。结果Lowry法的测定结果明显高于Bradford法,Bradford法测定结果与两种液相色谱方法相当。结论 Lowry法由于赋形剂的干扰,测定结果偏高,不宜采用;柱前衍生HPLC法虽然灵敏度更高,但操作较繁琐;Bradford法和RP-HPLC法可作为注射用尖吻蝮蛇血凝酶蛋白质含量的测定方法。 |

| Abstract: | Purpose To select a suitable method for the determination of protein content in Haemocoagulase Agkistrodon for injection.Methods Four methods including Lowry′s,Bradford′s,RP-HPLC and 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate(AQC) pre-column derivatization HPLC were compared in determining the protein content of Haemocoagulase Agkistrodon for injection.Results The values of proteins content in Haemocoagulase Agkistrodon for injection by Lowry′s method were significantly higher than those by Bradford′s method,while the results of the two HPLC methods were almost equal to Bradford′s method.Conclusion Because of the interference of excipients,the results were much higher when using Lowry′s method.Although the AQC pre-column derivatization HPLC method was much more sensitive,but the operation is relatively complex.Both Bradford′s method and RP-HPLC method can be selected as the protein content determination method of Haemocoagulase Agkistrodon for injection. |
