miR-155及SOCS1蛋白在酒精性脂肪肝小鼠肝脏中的表达

摘    要:目的 探讨Lieber-DeCarli液体饮食诱导的酒精性脂肪肝模型小鼠肝组织中miR-155及细胞因子信号转导抑制物1(SOCS1)蛋白的表达变化.方法 选取C56BL/6雄性小鼠,随机分为对照组及模型组.采用Lieber-DeCarli液体饮食诱导小鼠酒精性脂肪肝模型.试剂盒检测血清丙氨酸转氨酶(ALT)及天冬氨酸转氨酶(AST)水平.HE染色、油红染色观察小鼠肝损伤变化.qRT-PCR法检测小鼠肝组织中miR-155的表达量.Western blot法检测小鼠肝组织中SOCS1蛋白的表达变化.采用Pearson检验分析miR-155与SOCS1蛋白表达的相关性.结果 病理结果及血清ALT、AST水平证实酒精性脂肪肝小鼠模型建立成功.qRT-PCR结果显示模型组小鼠肝组织中miR-155表达量明显高于对照组(P<0.001).Western blot结果显示模型组小鼠肝组织中SOCS1蛋白表达明显低于对照组(P<0.01).Pearson相关检验结果显示miR-155与SOCS1蛋白表达呈负相关性(r=-0.916,P<0.05).结论 miR-155及SOCS1蛋白可能在酒精性脂肪肝发病过程中起着重要作用,确切机制有待进一步研究.

The expression of miR-155 and SOCS1 protein in the liver of alcoholic fatty liver disease mice
Abstract:Objective To explore the expression of miR-155 and SOCS1 protein in the liver of alcoholic fatty liver disease mice induced by Lieber-DeCarli.Methods C56BL/6 male mice were used and divided into the control group and model group randomly.Lieber-DeCarli was used to induce alcoholic fatty liver disease model.ALT and AST level were detected by commercial kit.Histological analysis of mice liver was detected by HE and oil red staining.The expression of miR-155 in liver was detected by qRT-PCR,and the expression of SOCS1 protein in liver was detected by Western blot.Pearson correlation test was used to analyze the correlation between miR-155 and SOCS1 protein expression.Results Liver pathological changes and the levels of serum ALT and AST showed that alcoholic fatty liver disease mice model was established successfully.The expression of miR-155 in the model group liver was increased versus the control group(P <0.001),while the expression of SOCS1 protein in the model group liver was decreased versus the control group(P <0.01).Pearson correlation test results showed that miR-155 was negatively correlated with SOCS1 protein expression(r=-0.916,P<0.05).Conclusion miR-155 and SOCS1 may play a vital role in occurrence and progress of alcoholic fatty liver disease.
