MMP-14和CD31-MVD在结直肠癌组织中的表达及临床意义

目的 探讨基质金属蛋白酶-14(MMP-14)和CD31标记的微血管密度(CD31-MVD)在结肠癌组织中的表达及其与肿瘤的临床意义.方法 采用免疫组织化学方法检测60例结直肠癌患者的癌组织及20例正常切缘组织中MMP-14和CD31-MVD的表达.采用qRT-PCR的方法检测20例新鲜结直肠癌组织及正常切缘组织中MMP-14 mRNA的相对表达量.结果 ①结直肠癌组织中的MMP-14阳性表达率以及CD3 1-MVD值明显高于正常切缘组织(P＜0.05);②MMP-14的表达及CD31-MVD值与肿瘤大小、浸润深度、TNM分期和淋巴结转移有相关性(P＜0.05),而与患者性别、年龄无明显相关;③MMP-14的表达与CD31-MVD呈正相关性(P＜0.05);④结直肠癌组织中MMP-14 mRNA的相对表达量显著高于正常切缘组织(P＜0.01).结论 结直肠癌组织中MMP-14的高表达和CD31标记的肿瘤组织微血管密度与结直肠癌的发生、浸润、转移有密切关系.

Expression of MMP-14 and CD31-MVD in colorectal carcinoma and its clinical significance

Objective To investigate the expression of matrix metalloproteinase-14 (MMP-14) and CD31-MVD in colorectal carcinoma and their clinical significance.Methods The expression of MMP-14 and CD31-MVD were detected by immunohistochemistry in 60 cases of colorectal carcinoma and 20 cases of normal tissues from resection margin.The expression of MMP-14 mRNA were detected by qRT-PCR in 20 cases of fresh colorectal carcinoma tissues and normal tissues from resection margin.Results ① The expression of MMP-14 and CD31-MVD in colorectal carcinoma tissues were significantly higher than those in the normal tissues from resection margin (P ＜ 0.05).② The expression of MMP-14 and CD31-MVD were correlated with tumor size,depth of invasion,TNM stage and lymph node metastasis (P ＜ 0.05),but there was no significant correlation with gender and age of patients.③ The expression of MMP-14 was positively correlated with CD31-MVD.④ The expression of MMP-14 mRNA in colorectal carcinoma were significantly higher than that in the normal tissues from resection margin(P ＜ 0.01).Conclusion The high expression of MMP-14 and CD31-MVD in colorectal carcinoma were closely related to the occurrence,invasion and metastasis of colorectal cancer.
