益元活血丹胶囊对老龄大鼠脑缺血/再灌注一氧化氮和肿瘤坏死因子的影响

摘    要:
目的:从一氧化氮(NO)和一氧化氮合成酶(NOS)及肿瘤坏死因子(TNF)的变化方面研究益元活血丹对老龄大鼠脑缺血/再灌注损伤防护作用的机制,为防治脑梗塞提供实验依据。方法:老龄(20月龄以上)大鼠分为老龄对照组、老龄模型组、尼莫地平组、益元活血丹大、小剂量组、血栓心脉宁组,观察大鼠全脑缺血30 min 再灌注60 min 后血、脑组织中 NO 和 NOS 及脑组织 TNF 的水平。结果:与老龄模型组比较,老龄对照组、益元活血丹大剂量组、血栓心脉宁组血清中 NO 水平和益元活血丹大、小剂量组血清中 NOS 水平增高(P<0.05,P<0.01)。益元活血丹小剂量组脑组织中 NO 和 NOS 水平高于老龄模型组(P<0.05,P<0.01)。老龄模型组脑中 TNF 水平高于老龄对照组(P<0.01),尼莫地平组、益元活血丹大、小剂量组、血栓心脉宁组低于老龄模型组(P<0.05,P<0.01)。结论:NO 含量降低和 TNF 增高与老龄大鼠脑缺血/再灌注脑组织损伤有关。各组药物均能通过降低脑组织 TNF 含量以改善脑组织损害;益元活血丹能够通过提高血清和脑组织中 NOS 活性而促进 NO 的合成以拮抗脑组织损伤。

Abstract:
Objective:To study the protective effects of Yiyuan Huoxuedan Capsule (YYHXD) on cerebral ischemia/reperfusion injury in the aged rats by means of the changes of nitric oxide (NO),nitric oxide synthetase (NOS) and tumor necrosis factor (TNF) so as to pro- vide theoretical basis for the prevention and treatment of cerebral infarction.Methods:The aged rats (older than 20 months) were di- vided into six groups:normal control group,model group,nimodipine group,high and low dosage YYHXD groups,and Xueshuan Xin- maining (XXN) group.The blood NO level and NOS activity of blood and brain tissue,brain TNF level of rats with 60 min reperfusion after 30 min cerebral ischemia were performed.Results:The serum NO level in the aged model group was lower than that in the aged control group (P<0.05),and was lower in the aged model group compared with that in XXN and YYHXD groups respectively(P< 0.01-0.05).The NO activity in the high and low dosage YYHXD groups had a greater increase than that in the aged model group (P <0.01).The brain NO level in the aged model group had a decreasing trend in comparing to that in the aged model group,the NO lev- el of YYHXD was higher than that in aged model group (P<0.05).In comparing with aged model group,the TNF level in aged mod- el group was higher (P<0.05).The TNF level in nimodipine group,XXN group,low and high dosage YYHXD groups were lower than that in aged model group (P<0.01-0.05).Conclusion:The decrease of NO and increase of TNF were correlated with cerebral ischemia/reperfusion injury in the aged rats.YYHXD,XXN and nimodipine could alleviate the injury of brain by means of reducing the TNF level of brain.YYHXD could promote the synthesis of NO to prevent and treat the brain injury via improving NOS activity.
