自体骨髓间充质干细胞促进大鼠坐骨神经缺损修复的实验研究

摘    要:目的 观察自体骨髓间充质干细胞在神经再生室中的作用 ,探讨其作为组织工程的种子细胞修复周围神经缺损的可行性。方法 成年Wistar大鼠 2 4只 ,随机分成 3组 ,每组 8只 ,硅胶导管桥接右侧 13mm的坐骨神经缺损 ,A组在神经再生室内植入骨髓间充质干细胞 ECM (extracellularmatrix ,ECM )凝胶 ;B组植入ECM凝胶 ;C组注入生理盐水。术后 12周 ,进行大体观察及坐骨神经功能指数测定、电生理检测、神经组织学等观察。结果 A、B两组均有再生神经生成 ,但A组各项检测指标均优于B组 (P <0 0 5或 0 0 1)。结论 自体骨髓间充质干细胞能够促进周围神经再生 ,可以作为种子细胞应用于外周神经组织工程。

Abstract:Objective To evaluate the feasibility of using autologous mesenchymal stem cells as seed cells to repair peripheral nerve defects in a adult rat model. Methods Twenty-four adult Wistar rats were divided into 3 groups equally. A 13 mm-long nerve defect was made in the right sciatic nerve and was bridged by a 16 mm-long silicone tube. The silicone tube was filled with the compound of autologous mesenchymal stem cells and extracellular matrix (ECM) gel for group A, ECM gel for group B and saline for group C. Gross observation, electrophysiological examination, hisotological staining and measurement of nerves sciatic nerve function index were carried out at 12 weeks post-operation. Results At 12 weeks post-operation, regenerated nerve existed at both group A and group B. However, each mesurement index in group A was superior to that in group B (P<0.05 or P<0.01). Conclusion Autologous mesenchymal stem cells can promote peripheral nerve regeneration and can be used as seed cells for nerve tissue engineering.
