头针疗法治疗血管性痴呆大鼠的实验研究

摘    要:
目的探讨头针疗法治疗血管性痴呆的疗效及可能机制。方法采用国际公认4-血管阻断法制成血管性痴呆大鼠模型,将30只痴呆大鼠随机分成模型组、头针组、西药组。另取10只大鼠作为对照组。头针组取顶中线、顶颞前斜线、额中线、颞顶线,总共治疗30 d。药物组给予都可喜12 mg/kg每天1次,共治疗30 d。各组于30 d后进行学习记忆测试,腹主动脉取血测量其过氧化酶歧化物(SOD)、丙二醛(MDA)含量,取出双侧海马测量其乙酰胆碱酯酶(AchE)和生长抑素(SS)含量。结果模型组术后学习记忆能力严重障碍。头针组、都可喜组学习记忆能力明显改善(P<0.01);模型组术后海马AchE、SS较对照组明显减少(P<0.01),头针组、都可喜组与模型组比较两者明显升高(P<0.01)。模型组血浆中SOD活性降低,MDA活性升高,头针组、都可喜组与模型组比较2种活性有显著差异(P<0.01)。结论头针疗法能改善VD大鼠行为记忆能力,并改善海马区大量神经元多种神经递质指标,为其有效性提供实验依据。

Abstract:
Objective It is to explore the curative effect and possible mechanism of head acupuncture on rats with vascular dementia.Methods 30 cases of rats which were made into vascular dementia models successfully were divided randomly into model group,head acupuncture(HA) group and western medicine group.Another 10 rats were chosen as control group.HA group was treated with HA on top temple line,middle forehead line and so on for 30 days.Medicine group was given Dukexi 12 mg/kg once every day and for 30 days.Then the learning and memory abilities of all the rats in every group were tested,and their blood from abdominal aorta was gotten to measure the contents of SOD and MDA,hippocampus in both sides was gotten to determine the contents of AchE and SS.Results The abilities of learning and memory of rats in model group were bad,and those in HA group and Dukexi group were improved obviously(P<0.01).Compared with those in control group,AchE and SS obviously decreased in model group(P<0.01),and compared with the latter group,those increased significantly in HA group and Dukexi group(P<0.01).The activity of blood plasma SOD lowed down and that of MDA rose up in model group,and compared with those in HA group and Dukexi group,the differences were significant(P<0.01).Conclusion HA can improve rats' abilities of action memory and indexes of kinds of neurotransmitter of many neurons in hippocampus,and that may be the experimental evidence for its efficacy.
