己酮可可碱和苯那普利联合应用对阿霉素肾病大鼠细胞因子的影响

摘    要:目的 探讨己酮可可碱和苯那普利联用对阿霉素肾病大鼠细胞因子及尿蛋白的影响.方法 80只雄性SD大鼠,分为对照组、模型组、己酮可可碱组、苯那普利组及联合组.对模型组和三治疗组给予阿霉素(5 mg/kg) 尾静脉注射,建立肾病综合征模型.自注射阿霉素后,己酮可可碱组和苯那普利组分别给予己酮可可碱50 mg/(kg*d)、苯那普利6 mg/(kg*d)、联合组给予己酮可可碱25 mg/(kg*d)联合苯那普利3 mg/(kg*d)灌胃,共6周.分别于给药2周、4周、6周抽血测肿瘤坏死因子-α、白介素-6及放入代谢笼留24 h尿测蛋白定量.结果 肿瘤坏死因子-α、白细胞介素-6在己酮可可碱组、苯那普利组和联合组均较模型组明显降低(P均<0.01),而联合组又低于己酮可可碱组和苯那普利组(P均<0.05).结论 己酮可可碱和苯那普利联合应用能进一步降低肿瘤坏死因子-α和白介素-6的表达,减轻炎症反应,联合治疗疗效优于单独应用.

Abstract:Objective To investigate the effects of the combination treatment with pentoxifylline and benazepril on cell factor and urine protein in the kidney of adriamycin nephritic rats.Methods Eighty male Sprague-Dawley rats were divided into control group,model group,pentoxifylline treated group,benazepfil treated group and co-treated group,The rats from later four groups were injected with adriamyein(5 mg/kg) through tail vein,in order to set up the model of nephrotic syndrome.The rats from later three treated groups were respectively gastric-perfused with pentoxifylline 50 mg/(kg·d),benazepril 6 mg/(kg·d),both pentoxifylline 25 mg/(kg·d) and benazepril 3 mg/(kg·d) every day for 6 weeks.THe contents of TNF-a,IL-6 and pnoteinuria in all rats wene determined after injecting 2,4,6 week.Results A significant increase of TNF-a and IL-6 in pentoxifylline treated group,benazepril treated group and co-treated group compared with those in the model group(P<0.05),while that in co-treated group was lower than that in pentoxifylline and benazepril group(P<0.05,respectively).Conclusion The combination of the treatment with pentoxifylline and nazepri1 can more reduce the serum TNF-α and IL-6 level,decrease inflammatory reaction.The effect of combination treatment with pentoxifylline and benazepril is superior to monotherapy.
