贵州水族人群D13S317、D7S820和D16S519 STR Loci遗传多态性

摘    要:目的:调查贵州水族人群D13S317、D7S820和D16S519 STR基因座遗传多态性。方法:采用Chelex-100快速提取法提取贵州省荔波县水尧水族乡44名水族无关个体EDTA抗凝血标本DNA,再通过PCR复合扩增,变性聚丙烯酰胺凝胶电泳和银染分型,最后用Modified—Powerstats统计软件包输出各群体遗传学数据。结果:贵州水族人群D13S317,D7S820和D16S519基因座分别检出7个、6个和6个等位基因,各基因座基因型分布均符合Hardy—Weinberg平衡定律(P〉0.05)。D13S317,D7S820和D16S519基因座的杂合度分别为0.636,0.818和0.818;个体识别律分别为0.909,0.893和0.876;多态信息量分别为0.74,O.75和0.75;非父排除率分别为:0.337,0.633和0.633。结论:贵州水族D7S820和D16S519两个基因座有较高的遗传多态性,在法医学的个体识别和群体遗传学的研究中有较高的应用价值。

Abstract:Objective: To investigate the genetic polymorphism on STR Loci ( D13S317, D7S820 and D16S519) of Shui population in Guizhou Province. Methods: Using the Chelex - 100 fast extraction, we extracted EDTA anticoagulant blood specimen DNA from 44 unrelated individuals of Shui nationality in Shuiyao of Libo county in Guizhou province; and then amplified by the PCR; and typed with denaturing polyacrylamide gel electrophoresis and silverstaining; finally output the genetics data of each population with Modified - Powerstats software. Results: STR loci ( D13 S317, D7 S820 and D16S519 ) of Shui population in Guizhou province were detected 7, 6 and 6 alleles, whose genotype distribution rule conformed to the Hardy - Weinberg balance rule (P 〉 0. 05) . And the heterozygosity (H) of D13S317, D7S820 and D16S519 are 0. 636, 0. 818 and 0. 818; the discrimination power (DP) are 0. 909, 0. 893 and 0. 876; polymorphism information content (PIC) are 0. 74, 0. 75 and 0. 75 ; and probabilities of paternity exclusion (PPE) are 0. 337, 0. 633 and 0. 633. Conclusion: There are greater genetic polymorphism on D7S820 and D16S519, which is valuable for individual discrimination of forensic medicine and the study of population genetics.
