摘 要: | 目的:探讨梅毒患者外周血T细胞表观遗传学DNA甲基化状态和组蛋白修饰水平检测的意义。方法:选取我院收治的一期梅毒、二期梅毒、梅毒血清学固定患者各10例为观察组,10例正常志愿者为对照组,比较各组患者外周血T细胞基因组DNA甲基化水平、组蛋白甲基化和乙酰化修饰的表达水平。结果:四组外周血细胞DNA甲基化水平分别为(19.70±4.13)%、(13.62±3.46)%、(9.31±2.82)%、(26.34±6.35)%,组蛋白H3K9甲基化水平分别为(0.324±0.124)、(0.210±0.130)、(0.132±0.124)、(0.677±0.128),组蛋白H3乙酰化水平分别为(0.164±0.024)、(0.103±0.010)、(0.052±0.014)、(0.211±0.012),各组比较差异均有统计学意义(F=28.43,F=36.21,F=190.35;P<0.05),对照组水平最高、梅毒血清学固定组水平最低;四组外周血细胞组蛋白H3K4甲基化水平有渐减的趋势,但差异无统计学意义(F=2.07;P>0.05)。组蛋白H4乙酰化水平无明显变化,差异无统计学意义(F=0.04;P>0.05)。结论:T细胞DNA低甲基化、组蛋白H3K9甲基化和H3乙酰化参与梅毒患者的细胞免疫功能的调控,为防治梅毒提供新的思路和干预手段。

Abstract: | Objective: To discuss the significance of epigenetics DNA methylation state and histone modification in peripheral blood T cells of syphilis patients.Methods:Ten patients with primary syphilis,10 with secondary syphilis and 10 with syphilis serological fixed were selected as the observation group,at the same time 10 healthy volunteers were also enrolled as control group.The expression level of epigenetics DNA methylation and histone modification in peripheral blood T cells were compared among different groups.Results:The levels of DNA methylation in peripheral blood T cells of these four groups were(19.70±4.13)%,(13.62±3.46)%,(9.31±2.82)% and(26.34±6.35)%,respectively,the levels of H3K9 methylation respectively were(0.324±0.124),(0.210±0.130),(0.132±0.124) and(0.677±0.128),the levels of histone H3 acetylation respectively were(0.164±0.024),(0.103±0.010),(0.052±0.014) and(0.211±0.012).Differences were all statistically significant(F=28.43,F=36.21,F=190.35;P<0.05).These levels were the highest in the control group,while lowest in the syphilis serological fixed group.The levels of histone H3K4 methylation of four groups showed a decreasing tendency,but the differences was not statistically significant(F=2.07;P>0.05).The levels of H4 acylation showed no significant changes(F=0.04;P>0.05).Conclusions:The DNA methylation,histone H3K9 methylation and H3 acetylation of peripheral blood T cells all involved with cellular immune function control of syphilis patients,which indicates a new method for prevention and control of syphilis. |
