HCCR1和HCCR2可溶性蛋白在细菌中表达、纯化及在早期肝癌诊断中的应用

目的评估HCCR1和HCRR2克隆、细茵表达及可溶性蛋白的纯化及应用于早期肝癌诊断的可行性。方法cDNA克隆、表达、蛋白质的纯化、Westem blot分析用于评估HCCR1和HCCR2蛋白的生产，Elisa方法用检测血清抗原。结果HCCR1和HCCR2能够高水平地在细茵中表达，纯化的可溶性蛋白能用于生产抗体并应用于肝癌的早期诊断。结论HCCR1和HCCR2蛋白质可以在细茵中生产，并能成功地用于肝癌的早期诊断。

Soluble recombinant proteins of HCCR1 and HCCR2 expression and purification in bacteria and its application for novel early diagnosis of hepatocellular carcinomas

[Objective] HCCR1 and HCCR2 are the oncogenes overexpressed in hepatocellular carcinomas(HCC).In this experiment,it was evaluated that the cloning,bacterial expression,purification and characterization of soluble recombinant proteins of HCCR1 and HCCR2 for novel early diagnosis of HCC.Protein solublity was key to producing recombinant proteins for founction determination.[Method] Five methods,cDNA clone and expression,protein purification,solubility assay and western blod assay were used to evaluate production of HCCR1 and HCCR2 proteins,enzyme-linked immunosorbent assay(ELISA)was used to detect HCCR1 and HCCR2 antigens in serum.[Result] The results show that the proteins of HCCR1 and HCCR2 were expressed in Escherichia coli at high levels.[Conclusion] The recombinant proteins are soluble and may used to generate the antibody for early diagnosis of HCC.Finally,HCCR1 and HCCR2 may be useful in monitoring HCC,and as potential disease specific therapeutic targets.
