| 摘 要： | 目的探讨心肌组织块体外原代培养豚鼠心肌细胞的方法。方法将剪成1mm3左右心肌组织小块,采用心脏组织块体外贴块种植原代培养豚鼠心肌细胞。结果该方法能成功地培养出原代心肌细胞,并能够传代,倒置相差显微镜下证实培养的细胞为心肌细胞,免疫组化染色鉴定为心肌细胞特异性蛋白cTnT表达阳性,同时具有心肌细胞节律收缩特性,膜片钳可以记录到细胞Ik及Ito电流。结论心肌组织块体外原代培养可以获得较多单个心肌细胞,所获得的细胞具有良好的功能状态,操作方法简单。 |

| Abstract: | Objective To evaluate the method of cardiac tissue explanted in myocytes isolation and culture.Methods The heart of guinea pig was isolated,and cut in small pieces.The cardiac tissue were cultured in medium.Results The single myocyte explanted from the tissue pieces.The cells can be passaged in 2-3 generation.The immunochemical stain of cTnT were positive in the cells.The Ik and Ito current could be successful recorded in patch clamp study.Conclusion By the method of cardiac tissue explantation,cardiac single myocyte can be obtained easily.The method of tissue culture is simple. |
