硫酸软骨素酶ABC促进大鼠脊髓损伤后神经功能恢复的实验研究

摘    要:目的 探讨硫酸软骨素酶ABC(ChABC)对神经脊髓损伤后运动功能恢复的影响.方法 SD大鼠72只,雌雄不限,随机分为假手术组、生理盐水对照组和ChABC治疗组,采用Allen法打击大鼠胸10脊髓损伤模型,分别在伤后即刻和随后每天一次连续一周蛛网膜下注射生理盐水和ChABC.HE染色和尼氏染色观察各时间点脊髓损伤组织形态和尼氏体及神经元的变化,采用BBB功能评分和运动诱发电位(MEP)观察大鼠的运动功能恢复情况.结果 大鼠脊髓损伤后1周时BBB评分和对照组无显著差别,在2、4周,治疗组评分结果明显优于对照组(P<0.05;P<0.01);MEP在1、4周的N1波潜伏期与对照组差异显著(P<0.05;P<0.01).HE和Nissl染色显示治疗组的形态和神经元数量要优于对照组.结论 ChABC能促进大鼠脊髓损伤后神经运动功能恢复,并对脊髓组织损伤具有保护作用.

Abstract:Objective To investigate the effect of Chondritinase ABC(ChABC)on recovery of locomotor function after spinal cord injury(SCI).Methods Seventy-two SD rats were randomly divided into three groups:sham group,saline group and ChABC group.The SCI model of contusion was established according to the method of Allen's and then the saline and ChABC group were treated every day for 1 week with saline,ChABC respectively by subarachnoid space catheter placed near the injury site.Motor behavior was measured by the method of BBB scores and Motor Evoked Potentials(MEP)were recorded to display the conduction of injured spial cord.The morphous of the remnant injured spinal cord tissue and the quantities of nuerons were detected by HE staining and Nissl's steining.Results There were no differences for BBB scors beteen the saline and ChABC group within 1 week,but from 2 to 4 weeks the BBB scors had significantly differences(P<0.05;P<0.01 respectively);and MEPs were significantly enhanced in ChABC group compared with saline groups(P<0.01 in 4 week);the morphous and the nuerons of the remnant injured spinal cord in ChABC group was better than in saline group.Conclusion The result indicate the ChABC can enhance the recovery of neuromotor function and protect the injured spial tissue.
