槐角不同部位中芦丁和槐角苷含量的比较研究

摘    要:目的考查槐角不同部位,即角皮和种子占槐角重量的百分比,及各组分中芦丁和槐角苷的含量差异,为科学、有效地利用现有槐角资源提供理论依据。方法采用高效液相色谱法同时测定角皮或种子提取物中芦丁及槐角苷的含量。HPLC测定条件:Diamonsil C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-1%冰醋酸-乙腈(57∶38∶5,V/V),流速1.0ml.min-1,检测波长260 nm。结果槐角角皮和种子占槐角重量的平均百分比分别为64%和36%。芦丁浓度在0.08~1.0 g.ml-1,槐角苷浓度在0.125~2.0 g.ml-1范围内,与峰面积呈良好的线性关系。槐角皮和槐角种子中黄酮类化合物HPLC谱型相似,但含量差别较大,槐角皮中芦丁与槐角苷含量分别为(0.44±0.15)%和(6.20±0.57)%,种子中两者的含量分别为(0.36±0.09)%和(0.49±0.20)%。槐角皮中槐角苷的含量明显高于槐角种子,P≤0.01,提示差异显著。结论槐角中的槐角苷主要存在于槐角皮中,因此提取这类化合物时可以将槐角皮和种子分开,只以角皮为材料,种子可用于其他目的的研究,以达到科学、有效地利用槐角资源。

Abstract:ObjectiveTo compare the differences in rutin and sophoricoside levels between pericarps and seeds of Sophora japonica L.MethodsRutin and sophoricoside were determined simultaneously by a method of RP-HPLC.The extracts prepared from pericarps and seeds of Sophora japonica L.were separated on a Diamonsil C18 column(150 mm×4.6 mm,5 μm).Methanol-acetonitrile-0.07% phosphoric acid(12∶20∶68,v/v) was used as mobile phase.The flow rate was 1.0 ml·min-1.The detection wavelength was at 260 nm.ResultsThe percentages of weight of pericarps and seeds in fructus sophorae were 64% and 36%,respectively.The linear ranges of rutin and sophoricoside were 0.08~1.0 g·ml-1 and 0.125~2.0 g·ml-1,respectively.The profiles of rutin and sophoricoside in pericarps and seeds were quite same,but the levels of them were different.The levels of rutin and sophoricoside in pericarps were(0.44±0.15)% and(6.20±0.57)%,respectively,and those in seeds were(0.36±0.09)% and(0.49±0.20)%,respectively.The level of sophoricoside in pericarps was much higher than that in seeds(P≤0.01).ConclusionThere are significant differences in the levels of sophoricoside between the pericarp and the seed of Sophora japonica L.,the fructus Sophorae can be used separately in order to be used effectively.
