MTDH基因沉默对B淋巴瘤细胞增殖及侵袭的影响

目的探讨异黏蛋白(MTDH)基因沉默对B淋巴瘤细胞增殖及侵袭能力的影响。方法在B淋巴瘤细胞Raji中转染MTDH siRNA和siRNA对照,分别命名为MTDH siRNA组和siRNA对照组,将没有转染的细胞作为对照组。qRT-PCR和Western blot方法检测细胞中MTDH表达水平,MTT方法检测细胞增殖和黏附能力,流式细胞术检测细胞凋亡,Transwell小室检测细胞侵袭及迁移能力,Western blot检测细胞中波形蛋白(Vimentin)、基质金属蛋白酶-2(MMP-2)、活化的Caspase-3(Cleaved Caspase-3)、基质金属蛋白酶-9(MMP-9)蛋白水平。结果MTDH siRNA细胞中MTDH mRNA和蛋白水平均明显低于对照组和siRNA对照组(P<0.05)。与对照组和siRNA对照组比较,MTDH siRNA细胞增殖和黏附能力降低,细胞凋亡率升高,细胞侵袭和迁移能力也降低,细胞中Vimentin、MMP-2、MMP-9蛋白水平降低,Cleaved Caspase-3蛋白水平升高,差异均有统计学意义(P<0.05)。结论MTDH基因沉默下调B淋巴瘤细胞增殖能力,抑制B淋巴瘤细胞侵袭和迁移。

Study on the effect of MTDH gene silencing on proliferation and invasion of B cell lymphoma

Objective To investigate the effects of MTDH gene silencing on proliferation and invasion of B cell lymphoma.Methods In B lymphoma Raji cells were transfected with MTDH siRNA and siRNA control,named MTDH siRNA and siRNA control,the non transfected cells as control.The expression of MTDH qRT-PCR and Western blot assay,MTT method were used to detect cell proliferation and adhesion ability,cell apoptosis was analyzed by flow cytometry.Transwell assay was performed to detect the cell invasion and migration ability,MMP-2,Vimentin,MMP-9 and Cleaved Caspase-3 protein level were detected by Western blot.Results MTDH mRNA and MTDH protein level in MTDH siRNA cells was significantly lower than that of Control and siRNA control(P<0.05).MTDH siRNA cells proliferation and adhesion reduced,apoptosis rate increased,the invasion and migration ability of cells also decreased,cells in Vimentin,MMP-2,MMP-9 protein level decreased,increased the protein level of Cleaved Caspase-3,compared with Control and siRNA control,the difference was statistically significant(P<0.05).Conclusion MTDH gene silencing reduced the proliferation of B lymphoma cell capacity,inhibit the invasion and migration of B cell lymphoma.
