三氧化二砷与刺五加合用诱导HeLa细胞凋亡的实验研究

目的探讨三氧化二砷与刺五加合用对癌细胞生长的抑制作用及其机理。方法以HeLa细胞的生长抑制情况,流式细胞仪PI单染法检测药物诱导细胞的凋亡状况。结果两药合用24 h组细胞变小变圆,脱落情况都较其他组明显,该组细胞生长抑制率明显高于单用组,合用24 h组细胞的凋亡率明显高于合用16 h和8 h组(P<0.01)。结论刺五加可以作为三氧化二砷的抗癌增效剂,两药合用有很强的抗癌活性,其作用机理与抑制癌细胞生长、诱导其凋亡有关。

Experimental Study on the Apoptosis of HeLa Cells Induced by Arsenic Trioxide and acanthopanax senticosus

ObjectiveTo investigate the anti-tumor effect of Arsenic trioxide and Acanthopanax Senticosus on carcinoma cells and its probable mechanism.MethodsHeLa cells were cultured as a pathological model.HeLa cells structural change were examined by inverted phase microscope.The inhibition effect on the growth of cells was evaluated by microculture tetrazolium assay(MTT).The apoptosis of cells were detected by flow cytometry(FCM).ResultsThe cells of the group of two medicines used together turned smaller and rounder than other groups.The growth inhibited rate of this group was higher than that of control group.The apoptosis rate of two medicines used together 24 hours was higher than 16 hours and 8 hours(P<0.01).ConclusionA-canthopanax Senticosus can improve the anti-tumor effects of Arsenic trioxide,but whether it can decrease the toxicities of Arsenic trioxide or not is worth more studying.
