金锁二仙饮对雄性小鼠肾精亏虚证治疗作用及其机理实验研究

目的：观察金锁二仙饮对恐伤肾小鼠的影响并探讨其补肾填精作用机理。方法：将60只小鼠随机分为正常组、模型组、金匮肾气丸组以及金锁二仙饮高、中和低剂量组,共6组。除正常组外,其余5组制造＂恐伤肾＂肾精亏虚动物模型,连续21天,造模第8天开始模型组每天灌胃生理盐水（0.2mL/10g）、金匮肾气丸组灌胃金匮肾气丸混悬液（2.7g·kg-1·d-1）、金锁二仙饮高、中和低剂量组分别以金锁二仙饮（36、18、9g·kg-1·d-1）灌胃,共14天。观察动物一般情况,体重、性腺重量及其指数,血清睾酮（T）、精子生成素（FSH）和间质细胞刺激素（LH）含量的变化。结果：金锁二仙饮能够改善小鼠肾精亏虚症状,增加小鼠体重及性腺重量,提高血清T、FSH、LH水平。结论：金锁二仙饮可能通过对下丘脑-垂体-性腺轴激素水平的合成和释放的调节,发挥补肾填精作用。

Therapeutic Effects and Mechanism of Jinsuo Erxian Yin on Kidney Essence Insufficiency of Male Mice

Objective：To observe the effects of Jinsuo Erxian Yin on the mice of kidney essence insufficiency and explore the mechanism of nourishing kidney essence. Methods： 60 healthy male Kunming mice were divided into 6 groups： normal group, model group, Jingui Shenqi Wan group, Jinsuo Erxian Yin high-dose, middle-dose and low-dose groups. Except normal group, other 5 groups were established the animal model of kidney essence insufficiency by the method of terror impairing kidney for 21 days. Starting from the eighth day, model group was given saline（0.2mL/10g）, Jingui Shenqi Wan group was given Jingui Shenqi Wan（2.7g·kg-1·d-1）, and Jinsuo Erxian Yin high-dose, middle-dose and low-dose groups were given different concentration Jinsuo Erxian Yin（36g·kg-1·d-1, 18g·kg-1·d-1, 9g·kg-1·d-1） respectively for 14 days. Then observed the general situation of animals, measured weights, gonad weights and the quantities of T, FSH, LH in serum. Results： Jinsuo Erxian Yin not only makes the symptoms of the mice of kidney essence insufficiency better, increases weights and gonad weights, but also improves the levels of T, FSH and LH in serum. Conclusion： Jinsuo Erxian Yin plays the role of nourishing kidney essence by regulating the synthesis and release of hormones in HPG axis.
