牛磺酸对大鼠脑创伤凝血状态影响的研究

摘    要:目的应用血栓弹力图(TEG)评价牛磺酸(taurine)治疗脑创伤(TBI)SD大鼠的血液凝固状态变化情况。方法 SD大鼠随机分为假手术组、TBI组、牛磺酸治疗组(TAU组)。TBI组和TAU组用液压脑损伤打击仪建立重型TBI模型(左脑打击)。造模成功后即刻从尾静脉给药,假手术组和TBI组给予相同剂量的生理盐水。牛磺酸治疗组给予牛磺酸治疗(给药量为200mg/kg)。每组动物连续给药7天,第8天从颈总静脉及腹主动脉取血,用TEG测量各组大鼠血液凝固情况。结果各组动脉血和静脉血比较:假手术组动脉血达到最大振幅的时间(TMA)、综合凝血指数(CI)与静脉血比较差异有统计学意义;TBI组动脉血反应时间(R)、血凝块的形成时间(K)、α角、最大振幅(MA)、CI与静脉血比较差异有统计学意义。各组静脉血组比较:TAU组静脉血CI值明显高于假手术组和TBI组。结论脑创伤8天大鼠的动脉血与静脉血液凝固状态存在差别,而TAU组则无此现象,这说明牛磺酸可能具有平衡TBI后动静脉血液凝固性的作用,但长时间使用牛磺酸,可能导致血液高凝状态。

Abstract:Objective Using thrombelastograph to study the change of blood coagulation in rats following acute severe traumatic brain injury after taurine treatment.Methods Thirty SD rats were randomly divided into 3 groups: sham group,traumatic brain injury(TBI) group and taurine group.The groups of TBI and taurine were subjected to severe traumatic injury of left brain by lateral fluid pressure device.Then rats in taurine group were injected taurine(dose:200 mg/kg) and the other groups were given saline through caudal vein immediately.The drug were given every day until the 7th day.The arterial blood from abdominal aorta and venous blood from jugular vein was drawn at the 8th day.Then blood coagulation was tested with thrombelastograph analyzer.Results Comparing arterial blood with venous blood,the differences of TMA and CI had statistical significance in sham group.The values of R,K,α angle,MA,and CI had statistical significance between arterial blood and venous blood in TBI group.In venous blood,the values of CI in taurine group were significantly increased compared with the sham group and TBI group.Conclusion After severe traumatic brain injury there was difference in blood coagulation between arterial blood and venous blood at the 8th day.But this phenomenon doesn't exist when treated with taurine,which implies that taurine may have the function of equilibrating the blood coagulation following traumatic brain injury,but it may also lead to blood hypercoagulation after the use of taurine for a long time.
