Abstract:Objective To explore the mobilization effects of recombinant human serum albumin-granulocyte colony-stimulating factor fusion protein(GW003) on hematopoietic cells in Rhesus monkeys. Methods Seven adult rhesus monkeys received a single injection of GW003 subcutaneously at a dose of 150 μg/kg. Peripheral blood cell counts and colony formation units(CFU)of peripheral blood(PB) were evaluated in the monkeys at different time after injection. Results Significant mobilization effects were observed in hematopoietic progenitors, leucocytes, neutrophils, monocytes and immature granulocytes of the healthy adult Rhesus monkeys. And PB cells counts peaked at 18 h after GW003 injection. At 10 h post-injection, the numbers of colony-forming peripheral blood cells such as CFU-GM, CFU-Mix and CFU-MK were increased 1.8, 1.9 and 1.4 fold, while BFU-E and CFU-E were increased 1.7 and 1.5 fold relative to those of the mokeys pre-injection. Conclusion GW003 has significant mobilization effects on PBS/PC, suggesting that it might be a novel and efficient mobilization agent for hematopoietic cell.
