摘    要:目的建立高灵敏度HPLC法测定大鼠血浆中昂丹司琼含量.方法大鼠血浆样本用乙酸乙酯液-液萃取后,采用Waters Atlantis C18(150 mm×4.6 mm,5μm)色谱柱,以奈韦拉平为内标,流动相为乙腈:0.002μL/mL磷酸二氢钾缓冲液(用磷酸调pH到3.3)=30:70,流速1.0 mL/min,柱温35℃,检测波长310nm.结果血浆中昂丹司琼2~1 000 ng/mL范围内线性关系良好,批内、批间精密度分别〈5.1%和7.7%,昂丹司琼提取回收率为90.4%~94.1%.结论 HPLC方法快速、灵敏、准确,适用于昂丹司琼血浆浓渡的测定及其药理实验研究.

Abstract:Objective To establish high-performance liquid chromatographic(HPLC)method for the quantification of ondansetron in rats plasma.Methods Sample preparation was accomplished through liquid-liquid extraction with ethyl acetate and chromatographic separation was performed on a reversed phase Waters Atlantis C18 column at 35 ℃.Mobile phase consisted of a mixture of acetonitrile-0.02 μL/mL KH2PO4 buffer(pH 3.3)(30:70,v/v)at a flow rate of 1.0 mL/min.Wavelength was set at 310 nm.Helicid was selected as the internal standard(IS).Results The linear range of ondansetron in plasma was 2~1000 ng/mL.The intra-day and inter-day precisions were lower than 5.1 and 7.7%,respectively.The recovery rate of ondansetron was 90.4~94.1%.Conclusion HPLC is rapid,sensitive and accurate,which can be used for determination of andansetron in rat plasma and its pharmacology study.
